| Biosecurity had become the key hamper that transgenic plant spreaded largely. In order to accumulate the scientific date for the biosecurity evaluation, the transgenic poplar with BtCry3Ac gene and transgenic poplar with BtCry1Ac gene were taken as the subject to study the effect of the two kinds transgenic poplar on the community composition, community character, and so on, by the field investigation and the resistance of transgenic poplar with BtCry3Ac gene against the target pest Apriona germari by laboratory expeiment. And the temporal and spatital dynamic expression of toxic protein was detected.The resusts were as follows.1 The study on the crown arthropods of transgenic poplar with BtCry3Ac and transgenic poplar with BtCry1Ac showed that the arthropods community stability was fewer than the CK poplar which could be disturbed by interior and exterior factors easily, while the stability of each functional group was more than the CK poplar; and the target pests of the two kinds transgenic poplar were all restrained, and the pest community stability was higher than CK poplar that coundn’t lead to pest eruption easily. And the temproal dynamic trend of the arthropods community was in substantial agreement among the two kinds transgenic poplar and CK poplar .2 The study on the soil arthropods of transgenic poplar with BtCry3Ac and transgenic poplar with BtCry1Ac showed that the arthropods community composition was more reasonable, and stability and quantity are much higher than CK poplar. There was no obvious difference in the temporal and spatial dynamic trend of three kinds of dominant species between transgenic and CK poplar.3 Six transgenic strains of of transgenic poplar with BtCry3Ac were selected to detected the stability of intergrated exogenous gene, the expression quantity of soluble protein and toxic protein. The results showed that the exogenous gene was stably integrated in the 6 transgenic strains’genomes. Toxic protein existed in xylem of each strain throughout growing season. The toxic protein in each xylem showed certain temporal pattern, all of which reached peak in August or September and started to drop quickly in October. Howere, the expression quantity was varied, among which strain CC84 (PCC1)had an obviously higher expression quantity over other strains from July to September.4 The identity results of transgenic poplar with BtCry3Ac gene resistance against Apriona germari showed that all 6 strains had certain inhibitory effect on oviposition of adult and growth of the larva of A.germari, showing certain resistance. Among the 6 strains, CC84 had a lethal effect of 50% to A.germari larvae and the resistance was fairly stable. Based on these identification results, it is suggested that transgenic strain CC84 has high potential for practical application.5 PCC1(CC84) of transgenic Btcry3Ac hybrid poplar 741 was further studied which resisted the pest stability,and the Bt toxic and soluble protein of xylem,leaf department and root segment were tested which were sampled at different development time and tissue . The results showed that there was no difference between transgenic and CK poplar in soluble protein content,but the soluble protein of the xylem and leaves of different part and the root presented a certain regularity that the expression of xylem and root part increased all the time and that the leaf part decreased then increased in the whole growing seasons. And the expression of Bt toxic protein in xylem increased from crown upper to lower and the expression of Bt toxic protein in leaf department decreased from crown upper to lower; the expression of Bt toxic protein in xylem increased then decreased and the peak time of expression was September, but leaf department and root segment did not appear a certain regularity with the time going forward . |
