Background: All the living things can resist stress from environment through signal transduction, to control the etablization process and mediate developmental process. Ca2+ ions play an essential role as second messengers in plant cells during various developmental processes and in response to environmental stimuli, such as abotic stress, light, pathogen and hotmone etc. Rice is one of the most important crop, and also a model plant for making research. Becterial blight which is caused by Xanthomonas oryzae pv. oryzae, is one of the serious rice disease. It can cause reduction and even zero yield. So, making research on resistant gene which mediated resistant response is necessary and significant. Calcineurin B-like proteins only exist in plant. They with CIPK(CBL-interacting protein kinase, CIPK) form one of the Ca2+ signal pathways. Now, there have been research shows that CBL family plays an essential role during developmental stages and resistance process.Aim: Produce antibodies of rice CBL. Understand the expression profile of OsCBLs during developmental stages and resistance process, to accumulate data for further research.Materials and Methods: Epitopes were predicted for candidate gene-encoded proteins and synthesized. Polyclonal antibodies were generated by rabbit immunization. Western blotting analysis was carried out for rice material collected at different developmental stages, further we examined their expression at different time points in the Xa21-mediated resistance against bacterial leaf blight.Results: Western blotting analysis was carried out to detect expression profile of 10 OsCBLs during developmental stages and Xa21-mediated resistance against bacterial leaf blight. 5 proteins were found related with the resistance response, they are OsCBL1、OsCBL3、OsCBL5、OsCBL9 and OsCBL10. Compare their expression in both the resistance and susceptibility response, discovering differential expression.Conclusion: Our research uncovered the expression profile of rice OsCBLs during developmental stages, and Xa21-mediated resistance against bacterial leaf blight. We found that OsCBL5 expressed specifically during seeding period, which indicated that it may play a role in seeding period. Our research also uncovered that most of the changed OsCBL proteins play positive roles during resistance response. Among 5 changed proteins, 4 up-expressing proteins play positive roles during resistance response; 1 down-expressing protein play negative roles during resistance response.Through comparing both of the resistance and susceptibility response, we found that the 5 changed OsCBLs were not induced by wound. OsCBL1 and OsCBL10 expressed differently between both of the resistance and susceptibility response, which indicated that they were specific protein during resistance response.Our research provided information and basis for uncovering functions of CBL during developmental stages and resistance process of rice.
