Studies on lily hybridization were more concentrated on wild Lilium spp.or the wild species and cultivars. However, hybridization of lily cultivars, especially Asiatic lily cultivars was less concentrated. In this study, lily cultivars of Asiatic hybrids and Oriental hybrids were employed as experimental materials for hybridization. To some extent, it can reflect cross-compatibility among lily cultivars and offer help for further study on lily hybridization.There were several contents in this study including the test of lily pollen vitality, hybridization, the observation of pollen tube growth by the fluorescence microscope, embryo rescue and the identification of hybridization and five hybrids were obtained ultimately. The main conclusions are as follows:1) The best culture media for pollen germination was sucrose 100g/L+H3BO340mg/L+CaCl2 10mg/L by orthogonal design. The pollen germination rates of’Black Out 1106’,’Detroit’,’Gironde’, ’Loreto’,’Mount Duckling’and’Tresor’were high while the germination rate of other cultivars were low.2) Most of the intraspecific crosses were compatible and could get swelling capsules. However, the interspecific crosses were incompatible. Although some of them obtained swelling capsules, the ovaries were small and the longevity of fruit was very short. It was also found that the results of cross and reciprocal cross were diverse for the given tow parents. For most of the crosses, the degree of capsule swelling and the embryo rate was positively correlated.3) It was observed under fluorescence microscope that in the crosses of’Mount Duckling’ self-pollination and’Loreto’×’Black Out 1106’, the pollen of’Mount Duckling’and’Black Out 1106’ could germinate on the stigma of’Mount Duckling’and’Loreto’respectively, and the pollen tubes could penetrate the styles, reach the basal of the style and finally arrive at the ovule in ovary. However, some incompatibility of the pollen tubes was observed in the growth process, such as.a series of callose responses in the style and ovary.4) The results suggested that the best embryo culture in vitro was MS+sucrose 3%+NAA 0.01mg/L for the embryo of’Navona’×’Tresor’. It was also observed that activated charcoal could promote rooting and the best rooting culture media for’Navona’×’Tresor’hybrid plantlets was MS+sucrose 6%+NAA 0.1 mg/L+activated charcoal 1.0g/L.5) ISSR-PCR was evaluated for its potential use in the identification of the five hybrids. Of the 15 ISSR primers subjected to screening,2 primers were selected for ISSR-PCR. Using these primers,29 discernible DNA fragments were generated with 24 (82.76%) being polymorphic bands.The result indicates that most DNA fragments of hybrids are the same with parents. The hybrids have two parents’ characteristic spectrum or one parent’s DNA fragments. Meanwhile, some hybrids show new DNA fragments that their parents do not have. The molecular evidence indicates the five hybrids are real hybrids.
