Immunohistochemical ultrasensitive SP method has been used to examine the expression of Bax, Bcl-2,Caspase-3andCaspase-9in the control rats and injected with ZEA oil rats in the study.Method:30SD rats at Ten-week-old were randomly divided into two groups andsimultaneous feeding. Test group (n=15): treat with0.5mL5mg/kg i.p. of ZEA.Controlgroup (n=15): treat with equal saline. Euthanized at3,6,12,24and48h after treatment inorder to determine the expression of Bax, Bcl-2, Caspase-3and Caspase-9at different times.Result:1. Copy of the pathology model on rats zearalenone intoxication Rats mainlyshow following symptoms: labia swelling, spirit weidun, loss of appetite, etc. Pathologicalchanges are expressed as follows: liver congestion swelling and texture crisp,lymphadenopathy, bleeding points on thymus, kidney congestion, Lung bleeding and becomered, tubal uterine edema, ovarian hyperemia. histopathological change:Control group werefound degeneration in liver cell and dilation of lobule central vein which were full of redblood cells; degeneration in the epithelial cells and luminal stenosis of the proximal tubules;plenty of megacells and hemosiderins in dilative splenic sinus; angiotelectasis in smallintestinal inherent layer and submucosa, tons of red blood cells in capillary vessel and visiblemacrophages infiltrates in tissue space; follicles in ovarian tissue were atretic.apotosis cellwere found in granular layer of ovarian follicle. By MG&P stain and TUNEL method, it isconfirmed that ZEA can result in apotosis of granular layer cell of ovarian follicle.2. The expression of Bcl-2and Bax the expression of Bcl-2and Bax in both experimentgroups and control groups and presented dynamic changes as time went on. Bax expressionshowed a significant increase compared to control groups (P﹤0.05) in3h,6h,12h andshowed steady decline at24h and48h. The expression of Bcl-2showed no statisticalsignificance compared to control groups(P﹥0.05).3. The expression of Caspase-3and Caspase-9Caspase-3expression showed asignificant increase compared to control groups (P﹤0.05) in3h,6h,12h,24h,48h. Theexpression of Caspase-9showed a significant increase compared to control groups (P﹤0.05) in3h,6h,12h and showed no statistical significance compared to control groups at24h and48h(P﹥0.05)..Conclusion: Zearalenone mycotoxicosis can result in ovarian follicles damage andgranulosa cell apotosis in rats.
