Taking the freeze-dried walnut green husk powder as the research object of the test, apreliminary exploration was conducted on the extraction process of its effective chemicalcomponents, the initial separation of its active substances, the contents of its chemicalcompositions of walnut green husks picked in different time, and the antifungal activity oftrichophyton rubrum from the different extracts. The results were as follows:⑴Carry out the analysis of the experiment, to obtain the optimum conditions forextracting the effective active ingredients of walnut green husk by the supersonic methodtogether with ethanol. The results showed that the volume fraction of ethanol was85%, theextraction time was55min, the extraction temperature was35°C, the number of extractiontimes was3, and the sample-solution ratio (mg·mL-1) was1:10.⑵Abstract the effective active ingredients of walnut green husk by the supersonicmethod together with ethanol, and then extract them with different organic solvents. Theresults showed that there was a significant difference in the contents of ethanol extraction andvarious extracts in the walnut green husks picked in different time, and the descending sizeorder of the extraction rate was August, July and June. The extraction rates of ethanol extract,petroleum ether extract, ethyl acetate, chloroform extract and raffinate aqueous phase inwalnut green husks picked in August were31.4%,7.10%,1.75%,2.15%and18.30%,respectively. Taking the residue of walnut green husks picked in different time after ethanolextraction process as the object of study, the supersonic method was adopted to assist inextracting the crude fiber and polysaccharides in the walnut green husk. The acid-base alcoholether method was used for extracting the crude fiber, and the results showed that the contentsof the crude fiber varied with the picking-time, and increased with the extension of thepicking-time. The extraction rates of the crude fiber in walnut green husks picked in June,July and August were18.09%,24.62%and27.47%respectively; The extration of the crudepolysaccharides in the green husk residue, and the results showed that the standard curveequation of the glucose was Y=0.01175+0.00903X, R=0.99909, and the contents of crudepolysaccharides changed different with the picking-time and decreased with the extension ofpicking-time. The extraction rates of crude polysaccharide in walnut green husks picked in June, July and August were31.90%,21.57%and12.60%, respectively.⑶The ultrasonic method was used to assist in extractingthe tanning matter in thewalnut green husks picked in different time, and the Folin-Denis colorimetric method wasused for its determination. The results showed that the standard curve of the tannic acid wasY=0.02486+0.00746X, R=0.99916, and the tannin contents of walnut green husks picked indifferent time were significantly different. The tannin contents of walnut green husks pickedin June, July and August were3.34%,4.16%and4.92%respectively.⑷Taking the walnut green husk picked in August as the object of study, extracted thealcohol extract of walnut green husk with different organic solvents, and then determined theinhibition effect of different extracts on the trichophyton rubrum. The results showed that allkinds of extracts from the walnut green husk had a certain inhibition effect on trichophytonrubrum. When the extract concentration of the walnut green husk organic solvent was40.0mg·mL-1, the inhibiting rates of petroleum ether extract, chloroform extract, ethyl acetateextract and raffinate aqueous phase was14.9%,42.4%,36.1%and35.6%respectively, andthe mycelial growth EC50was10.7100,5.3964,6.7345and6.3345mg·mL-1respectively. Theantifungal test on the extracts of different concentrations showed that, the greater theconcentration of the walnut green husk extract was, the better the antifungal activity would be.The MIC of the chloroform extract, ethyl acetate extract, raffinate aqueous phase andpetroleum ether extract on the trichophyton rubrum was respectively larger than5,8,10and15mg·mL-1.
