| Pepper(capsicum annuum L.) is an often cross-pollinated plant, and has redominantheterosis. The use of CMS can cut breeding cost, improve breeding efficiency and improveseeds purity. Especially the use of cytoplasmic male sterility(CMS) can facilitates theheterosis utilization due to the three lines system, avoiding the defect of remove50%fertileplants while use the AB line. Nevertheless, restorer lines mainly exist in the group of cayennepepper andred pepper, while bell peppers type Rf genes is hard to screen, which limits theuse of CMS due to the absence of the restorer lines. Therefore, research on the pepper malesterility mechanism is conductive to find out the pepper fertility restoration law which is ofgreat significance to the use of CMS in pepper breeding. It’s already achieved many researchresults in the period and manner of abortion, the discovery of the mitochondrial chimeric gene,mRNA editing et al by methods of cytology, physiology, biochemistry, biotechnology. Evenso, little research has been made on the differential proteome of anthers of CMS and thecorresponding maintainer line.CMS line FS1030A is used to identify the abortion period through the cytologyobservation by transmission electron microscope (TEM) and then to extract the antherproteome at the abortion stage.After purification, desalting treatment,2D-DIGE technologywas used for protein separation, Gel maps were analyzed by Decyder6.5(GE)software.Thirteen differential protein spots were identified by MALDI-TOF/TOF-MS. Mascot softwareis used to seek for ncbi-green plant protein database for protein biological information scienceanalysis, main results are as follows:1. The abortion occurred before tetrads came into being,when the heart leavessemi-expanded and the calyx closed. Two kinds of main structural changes in the tapetumwere observed in CMS FS1030A. First during the meiosis of microspore mother cells(MMCs),some of the tapetal cells were enlarged, elongated and developed into multilayer cells. Theysqueezed the MMCs excessively which narrow the powder chamber much. The MMCsdegenerated, vanished and failed to develop into tetrads, eventually leading to microsporeabortion. Second some of the premature tapetal cell layer degenerated, and then the remnantsof the collapsed tapetum and crushed MMCs formed a visible dense belt in the anther locule.The MMCs presented irregular shapes, and eventually degenerated due to the lack of nutrition.2. By Decyder6.5(GE) software analysis, approximately1070relatively distinct proteinspots were identified within pI3~10.A total of13differentially expressed protein spotswere found, with six upregulated and7downregulated in FS1030A. Use theMALDI-TOF/TOF-MS peptide fingerprint analysis, combined with the Mascot softwareretrieval to ncbi-green plant database. The identified proteins respectivel are:beta1,4N-acetylglucosaminyltransferase, putative histone H3protein,histone H1, ribosomal proteinL31, Agamous-like MADS-box protein AGL1,60S ribosoma lprotein L13a, AS-ZmSLRprotein,Putative chloroplast RNA processing protein, Ran-binding protein1b (RanBP1b),EMB2746(EMBRYO DEFECTIVE2746), maturaseK, pollenless3and one unknownprotein.They are involved in life processes which pepper the pepper CMS FS1030Aabortion may be related to, such as protein post-translational modifications, regulation of geneexpression and gametophyte development regulation and cell division control et al. |
