Alfalfa(Medicago sativa L.)belongs to the perennial leguminous cross-pollination herbaceous plants,which is monoecious and a female more than male,and removing stamen is the key to heterosis utilization.Compared with traditional way of artificial emasculation breeding,male sterility breeding has low cost,and little pollution.Theoretical study of alfalfa nuclear male sterile line has made some progress,but research for cytoplasmic male sterileneeds to develop further.Based on international and domestic research on economic crops mature technology for study of the cytoplasmic male sterile line and its homogeneic maintener line of alfalfa is significant.In this study,maintainer line MS-GN1 A and sterile line MS-GN1 B were analyzed for morphology cytology features as well as physiological and biochemical level,the main experimental results were as follows:Themaintainer line MS-GN1 A and sterile line MS-GN1 B continued to choose male sterility thoroughly,infertility with high degrees of strain for continuous generation of backcross,fertility,yield traits,quality traits,which analyzed of the innovation incubation male sterility,compact plant type of new cytoplasmic male sterile line material MS-GN1 A and its maintainer line MS-GN1 B.In this experiment,the five development periods of anther were observed under optical of Nikon-E200 microscope.The results showed the pollen grains of the maintainer lines were full,and cytoplasmic male sterile lines presented a dry state,and smaller and there were missing and fewer.In this study,we found that the cytoplasmic male sterility of alfalfa was the anomaly of the tapetum and the abortion of pollen during the tetrad.Compared with the retention system,the callosity of alfalfa sterile line did not dissolve the microspore.The pollen abortion of alfalfa sterile line has relationship with the abnormal development of the vascular bundle.In order to reveal the alfalfa physiological and biochemical mechanism of cytoplasmic male sterility,the different periods of bud of alfalfa male sterile(MS-GN1A)and alfalfa male fertile(MS-GN1B)were selected to analyze and compare the nutritional ingredients,including soluble sugar,starch,soluble protein and free proline,malondialdehyde content and antioxidant enzyme(CAT?POD and SOD).The results showed that the content of soluble sugar,starch,soluble protein and free proline were compared and contrasted deficit,and as an index of membranous peroxide malondialdehyde and activity of antioxidant enzymes were higher than MS-GN1 B.
