With the improvement of living standards and the modern human take more and more of fatty foods in their daily diet. This makes the incidence of fatty liver and hyperlipidemia increased substantially, and this cause the growing focus on research. Animal models have an important role to explore the pathogenesis and evaluation of fatty liver and hyperlipidemia so more and more animal models for fatty liver in the pathogenesis and drug prevention have established for a variety of experimental study in domestic and abroad.Chinese medicine culture is profound and traditional Chinese medicine and the active ingredient extracted from Chinese herbal medicine for study to become a research priority.The research to EGCG and resveratrol get the attention of scholars because of its special pharmacological effects to the disease and more and more in studies in humans and animals of this type of natural medicine have taken for nearly half a century.In this study, Male SD rats were fed with high fat diet for hyperlipidemia model.Subsequently, the model rats were divided into high-fat group, and EGCG group and resveratrol group two experimental groups and a control group with normal diet After4weeks administration, we observe the effect of these two drugs on rat fat weight, liver weight, liver coefficient, body fat and lipids and the degeneration of the liver tissue by light microscopy. We use Realtime-PCR to measure the expression of angiopoietin-like protein genes in liver and analysis the relationship between gene expression and this two dugs effection.Experiment I:Establishment of rat model of dietary fat. According to records, we establish fat rat model by fed high fat diet. After a week of modeling, the body weight and liver weight and liver coefficient of HF group is significant increased, There were significant differences compared to the normal group. The liver biopsy revealed lipid vesicles in the hepatic lobule. This prompt that high-fat model has been successfully established and follow-up experiment can carried out.Experiment II:The study of lipid-lowering effects of EGCG and resveratrol on high-fat model. Respectively to the two drugs to deal with the rats by oral gavage30mg/kg/d EGCG and30mg/kg/d of resveratroland normal group and model group deal with the same dose of distilled water for a month. At the end of the experiment,we weigh each rat body weight. Orbital venous blood and analysis the lipid. separate liver, epididymal fat, retroperitoneal fat and mesenteric fat and weight, calculation of the liver coefficient and body fat ratio.We cut each rat hepatic lobule and preserve in10%neutral buffered formalin and biopsy detection of liver lipid deposition.Result:Compared with the normal control group,the epididymal fat, retroperitoneal fat and mesenteric fat weight of model group is significantly increased. Compared with the model group,the epididymal fat, retroperitoneal fat and mesenteric fat weight of drug group is significantly decreased. Compared with the normal control group, the TG、TC、LDL-C of the model group is significantly increased but the HDL-C is significantly decreased. Compared with the model group, the TG、TC、LDL-C of the drug group is significantly decreased but the HDL-C is significantly increased.Light microscopic examination results show that steatosis of the liver in rats given EGCG and resveratrol treatment significantly reduced.Experiment III:The effect of EGCG and resveratrol on the expression of rat fat fatty liver lipid metabolism related genes.Angiopoietin-like protein mRNA expression changes in each group rat liver by extracting total RNA and reverse transcription PCR of cDNA and using Real-time quantitative PCR method for detection of rat liver. The results show that Angpt13expression levels in high-fat group significantly increased compared to the normal group and has statistically significant.EGCG can significantly reduce the high-fat group Angpt13expression with significant difference.Angptl4expression in high-fat group compared to the normal group does not have significant differences, but the EGCG group was significantly lowed than the high-fat group Angptl4expression with significant difference. The expression level of Angptl6in High-fat group was significantly decreased compared to the normal group and with significant difference. EGCG group expression and high-fat group compared to a rise.The Angpt13expression of high-fat group increased significantly than the normal group, resveratrol can significantly reduce the fat rat liver Angpt13expression with a significant difference.The expression does not have significant differences in amount of the Angpt14in the high-fat group and the normal group, resveratrol can significantly lower the Angptl4expression in high-fat group with significant difference. Angptl6expression in high-fat group decreased significantly than the normal group. The expression levels of Angptl6in resveratrol was significantly increased.
