The study was conducted to reveal the mRNA expression of GRa, Hsp70, Hsp90, GABAAα1Rand GABAB1R in different tissues of gilts during late gestation in hot summer. The object of this study was to provide the basic reference data for the molecular mechanisms of GABA used to mitigate stress.Twelve gilts (Landrace x Large White) during late gestation were assigned into two treatments (6gilts/treatment) according to the gestational day (90d and110d). The gilts were slaughtered and tissues including brain, ovary, heart, liver, spleen, lung and kidney were collected aseptically over liquid nitrogen and frozen at-80℃. Then total RNA isolation and reverse transcription, quantitative real-time PCR(qRT-PCR) was performed to determine the mRNA expression of GRa, Hsp70, Hsp90, GABAAα1R and GABAB1R in different tissues of gilts during late gestation. The results showed that the mRNA expression level of each gene in different tissues of gilts at d90of gestation was as follows:GRa:lung> spleen> liver> kidney> brain> ovary> heart; Hsp70:lung> spleen> liver> brain> kidney> ovary> heart; Hsp90: lung> kidney> ovary> brain> spleen> liver> heart; GABAAα1R:brain> lung> liver> ovary> spleen> kidney> heart; GABAB1R:spleen> lung> brain> kidney> ovary> liver> heart. d110:GRa:spleen> lung> liver> kidney> ovary> heart> brain; Hsp70:lung> spleen> kidney> brain> liver> ovary> heart; Hsp90:lung> ovary> spleen> kidney> brain> liver> heart; GABAAα1R:brain> lung> spleen> liver> ovary> kidney> heart; GABAB1R:spleen> lung> kidney> brain> ovary> liver> heart. Compared with d90of gestation, the expression of GRa and Hsp90mRNA was significantly reduced (P<0.01) and Hsp70mRNA was reduced (P<0.05) in the lung of gilts at d110of gestation. In the brain, the expression of GRa and Hsp90mRNA was significantly reduced (P<0.01). The expression of GRa and GABAAα1RmRNA was reduced (P<0.05) in the liver. The expression of GABAAα1R mRNA had an evident downward trend, but there was no significant difference (P>0.05). There were no gestation stage dependent effects on GRa, Hsp70, Hsp90, and GABABiR expression in all the other tissues (P>0.05).Our results indicated that the mRNA expression of GRa, Hsp70, Hsp90, GABAAα1R and GABAB1R had tissue specificity during late gestation. The mRNA of GRa, Hsp70, Hsp90and GABAB1R was preferentially expressed in the lung and spleen, while the GABAAα1R mRNA was abundantly expressed in the brain. The mRNA expression level of each gene was the lowest in heart. Compared with d90of gestation, the mRNA expression of GRa, Hsp70, Hsp90, GABAAα1R and GABAB1R were reduced in the brain, lung and liver of gilts at d110of gestation. There were no gestation stage dependent effects on GRa, Hsp70, Hsp90, GABAAα1R and GABAB1R expression in all the other analyzed tissues.
