Pine wilt disease which is a devastating disease on pine trees is an important internationalrecognized quarantine disease. At present people has done a lot of research on its pathogen andpathogenic mechanism, but there isn’t an exact conclusion until now. This article proposed to look for anew breakthrough in pathogenesis by analyzing of the changing of microbiota in the diseased pinetree.In addition, the nematicidal activity of Metarhizium anisopliae Mf2strain was tested, laying thefoundation for using Metarhizium anisopliae as biological pesticide to control Bursaphelenchusxylophilus.In this research, bacteria form healthy and diseased15years old Pinus massoniana were isolated.The result shows that:1、These were a large number of bacteria appeared in the LB flat medium withthe diseased plant wood one day later, however, the health one had nothing.2、purified the bacteria and12strains were isolated. Extracted bacterial genomic DNA, identified the bacteria using the method ofmolecular appraisal and got the results that Pectobacterium cypripedi,i Enterobacterasburiae,Enterobacter sp..Metarhizium anisopliae is one of the most applied entopathogenetic fungi. In this study, onemedium was choose after comparison among four common media PDA、PPDA、SDAY and SMAYwhich is most suitable for vegetative growth and spore production of anisopliae strains Mf2. We testedthe insecticidal effectiveness of M.anisopliae to pine wood nematode. The results showed that the rateof growth of Mf2is highest and the diameter of colony after14days of Mf2is largest in the PPDAmedium. The diameter of colony after14days is76.00mm. Conidia production of Mf2is highest in thePPDA medium which is2.5times of the second highest one. M.anisopliae have high nematicide activityto pine wood nematode. The mortality after36hour is98.52%. Pine wood nematodes are completelydissipated after5days. M.anisopliae has a good application potential to control pine wilt disease.
