We take the off-site preservation forest9119B of Larix olgensis in Qinshan forestry centre in Linkou, Heilongjiang province as the materials, and analyze the growth variation of the dominant tree population and inferior tree population.In this study take the off-site preservation forest9119B of Larix olgensis which are in Heilongjiang Province Qingshan forest centre as the experiment material., using SNP molecular markers and combining to phenotypic growth traits to analysis of genetic diversity in Larix olgensis.It can be a true the phenotypic selection of plus tree, but also to understand the interaction effects between growth traitsdgene.It has directive significance to formulate rational breeding strategies and Reasonable mobile phone genetic resources.To explore the genetic4CL of Larix olgensis, the development of EST-SNP and EST map construction, candidate genes LD mapping lay the foundation.The main findings are as follows:There is no significant difference between the dominant and inferior tree population, while the rich variations exist in height, diameter and volume between the populations.The volume variation coefficient is the richest one, then is the diameter and the least is the height, the coefficient is46.24%,19.82%and12.25%respectively. These three coefficient of dominant population are12.98%、20.77%and49.36%while the inferior population are11.47%,18.79%and43.16%.The variations of the dominant tree population are1.51%、1.98%and6.2%more than the inferior ones at height, diameter and volume respectively.There are significant differences between the populations by the variance analysis, according to analysis, we select6families (856,859、563、552、567、864)、3are dominant tree population and3are inferior tree population. The average volume of these6families are0.0223m3,,0.0186m3,0.0168m3,0.0164m3,0.0162m3and0.0159m3more than the average value of dominant population while0.0182m3,0.0145m3,0.0127m3,0.0123m3,0.0121m3and0.0117m3more than the inferior ones.According to10%selection rate among the dominant tree population and inferior tree population, the variation gain are59.33%、65.18%、74.91%、40.64%、40.94%、45.34%respectively.The orthogonal design was used to optimize ISSR-PCR amplification system of Larix gmelinii(RuPr.)RuP, Through the deep analyze,a suitable SNP-PCR reaction system was established,namly50μl reaction system containig31.4μl ddH2O,5.0μl cDNA,4.0μl10×PCR Buffer,2.0μl dNTP (2.5mM),4μl primer (10pm/μl),0.4μl Taq (0.5u/μl),3.2μl Mg2+(2.5mmol/L).The temperature profile used for SNP-PCR was94℃for3min;followed by35cycles of94℃for30s;56℃for30s,and72℃for30s:and was terminated with a7min DNA extension step72℃. SNP marker has Proved the Provenance of Larix olgensis.Rupr has a wealth of genetic diversity from the molecular level, and the marker indicate that the genetic variation mainly exists within populations, smaller variation exists among population SNP Primers detected a total of178SNPs.It sequences successfully193EST sequencing which overall length is83569bp, averaging469.5bp found a SNP sites, the sequences successfully wasl69, It has sequenced a total of156EST sequencing,and the successfully are119EST sequencing,in which the percentage is76.28%in the dominant tree population while in the inferior tree population has sequenced a total of84EST sequencing,and the successfully are74EST sequencing, in which the percentage is88.1%.Using the software DNAMAN analysis the sequencing result found that there are178SNPs,126belong to transition type, accounting for70.79%of the total variance; with52belong to transversion type, accounting for29.21%of the total variance. There are124SNP sites and89are transition accounting for71.77%of the dominant tree population variance, has35transversion for28.23%of the the dominant tree population variation. While there are54SNP site,37have belong to transition type, accounting for68.52%of the inferior tree population variance, has17belong to transversion type, accounting for31.48%of the inferior tree population variance. No matter in whole or in the advantage in the group or the inferior group, the mutant type roughly in line, transition:transversion=7:3. In the transition type, A/G and C/T transition account for37.08%and33.71%respectively; some types of transversion G/C, A/C, A/T, and G/T respectively in7.87%,8.99%,7.87%and4.99%.
