Wild rice (Oryza Rufipogon Griff.), an important rice germplasm pool, comprises a considerable number of advantageous genes, including cold resistance genes. Therefore, it is more appropriate to explore mechanisms of cold resistance and then easier to cultivate cold hardiness rice applying RNA-seq.The experimental materials for cold tolerance detection comprise12hybrid rice combinations, measure relative conductivity of seedling leaves as well as seedling rate of plants. The subjects of transcriptome include93-11, Dongxiang Wild Rice, Chaling Wild Rice, Mixed Overwintering Singles. The major findings are as follows:1The heterosis of cold tolerance at seedling stage in rice was analyzed through the electrical conductivity and the seedling rate of12hybrids and their parents. The analysis showed that, the relative electrical conductivity in over-parent heterosis was below zero; and in mid-parent heterosis was between-37.27%~+21.02%, in which the seedlings rate mid-parent heterosis was-30.80%-+7.20%. The heterosis was a mid-parent heterosis. And we also see that in the cold hardiness japonica rice superior.2All the amounts of the data obtained from the four groups of rice were around six million reads after processing. It was shown that cold treatment(4℃、3days) resulted in widespread genetic regulation changes as to all four groups of rice, comparing these data from different groups. The up-regulation genes significantly outgrew the down-regulation genes. The number of up-regulation gene of93-11is2691, while the number of down-regulation gene is1652; moreover, the numbers of Dongxiang Wild Rice is2322,1503, respectively; Chaling Wild Rice is2372,1553, respectively; Mixed Overwintering Single Plants is1836,1501, respectively.After comparing these data of Dongxiang Wild Rice, Chaling Wild Ricd and Mixed Overwintering Single Plants with that of9311, it was shown that the different down-regulation gene numbers is799,894and778, respectively; the different up-regulation gene numbers is775,922and388, respectively. 3In order to find out the cold resistance genes of the two wild rice and the mixed overwintering rice, we have analyzed the gene expression pattern of these kinds of rice and their control,93-11. It is discovered that three genes:OS03G0266300、 OS03G0266900、 OS03G0267000, have been specifically up-regulated, the alpha-crystallin-related small heat shock protein has been enriched by41folds. Down-regulated are156genes, which are distributed in photosynthesis genes, ferredoxin reductase genes.
