Identification Of Genes For Cold Resistance At Seedling Stage In Dongxiang Wild Rice (Oryza Rufipogon Griff.)

Rice is a kind of temperature sensitive crop,which is vulnerable to low temperature. Disastrous weather, such as cold spell in later spring and persistent rainy days, can greatly influence the developing of rice causing yield reduction. Dongxiang wild rice (Oryza rufipogon Griff.), as the most cold tolerant wild rice in the world as so far, has a strong ability to resist low temperature in seedling stage and heading stage. It will be significant to identify cold resistant genes from Dongxiang wild rice by molecular biological techniques and genome resequencing for researching the mechanism of wild rice cold resistance and breeding cold resistance cultivars.In this study, Dongxiang wild rice,9311and a F2population was used to identify cold resistant genes by genome resequencing and molecular markers. The results were summed as follows:Appraising the cold tolerance of F2plants by overwintering in the wild,6markers were found linkage to cold tolerance genes on chromosome1,3and12by BSA (Bulked Segregate Analysis). In addition, a lingkage map with108SSR markers was constructed and the tillering leaf conductivity of F2plants was used as the index of cold resistance to identify QTLs. The two QTLs, qSCR3and qSCR11, were identified on chromosome3and chromosome11,explaining16.3%and19.2%of phenotypic variances respectively.A large amount of SNP and Short Indel data were detected through genome resequencing. According to the mapping rusults and annotation documents,35genes were screened from mapped areas, including transcription relative genes (BGIOSGA012829), signal transmission relative genes(BGIOSGA034373, BGIOSGA012688,BGIOSGA012794, BGIOSGA012823and BGIOSGA034971) and membrane lipid relative genes(BGIOSGA010577,BGIOSGA034929,BGIOSGA034952).
