The research work of this paper includes that the relationship between the endogenous phytohormones in diploid and triploid Siraitia grosvenorii change and growth dynamic, the impact of exogenous hormones on the diploid and triploid Siraitia grosvenorii growth characteristics and quality, the diploid’s activity of POD,EST isoenzyme expression and LEAFY homologousgen,which was aim at announcing the physiological and biochemical characteristics and hormonal regulation effect on the two different ploidy of Siraitia grosvenori’s fruit development and could provide guiding the high-quality fruit and cultivation of new species with theoretic foundation. The main results were shown as follows:1During the fruit development,the contents of IAA, ZR, ABA endogenous phytohormones in diploid Siraitia grosvenorii was markedly increased as compared with the triploid’s.The contents of ABA endogenous phytohormones was the highest which was far greater than the other three hormones in the whole growth period of two ploidy Siraitia grosvenorii There were a lot of differences trend of change in the content of IAA between two ploidy fruits in the whole growth period. At day0to24of rapid growth, the contents of IAA rapidly increased and after the6th day sharp declined in the diploid;but at first the contents of IAA declined slowly and thel2th day rised rapidly in the triploid, with the enlargement of the fruit.The contents of GA3in the diploid showed a descending tendency throughout the growing season,but in the triploid showed a rising tendency until the fruits of late-maturing, addition to decline at day0to6; the contents of GA3two ploidy fruits increased in summary were on the contrary. There were significant differences with the contents of ZR in the former, but in the medium-term later tended to follow similar. When the time was at0to24day, the diploid ZR with the formation of the seed increased rapidly at first and then decline rapidly. The triploid’s decreased slowly at first and then increased rapidly. There showed that change trend of ABA content among two ploidy fruits followed the same pattern.2This paper studied and analyzed separately POD isozymes and EST isozymes in the growth process fruits of diploid and triploid Siraitia grosvenorii. The result showed that there were a lot of differences between POD isozymes and EST isozymes of diploid Siraitia grosvenorii fruits and triploids’, which the POD isozymes was the largest differences on fruits development and the bands were different from the others. we isolated nine POD isozymes bands from diploid Siraitia grosvenorii fruits,which P7, P8and P9were two specific bands.And also isolated ten POD isozymes bands from triploid Siraitia grosvenorii fruits,which p3,p6,p9and p10were two specific bands of POD isozymes.It showed that there were three differences isozyme bands of two ploidy fruits,with P4,p3,p4. And P4bands had obvious effect on promoting the enlargement of the fruits.We isolated ten EST isozymes bands from diploid and triploid Siraitia grosvenorii fruits, which E1,E3,E7,E9,E10were three specific bands of EST isozymes from the diploid Siraitia grosvenorii fruits,and el,e3,e5,e6,e9,e10were four specific bands of EST isozymes from the triploids’. The EST enzymes activity showed a general trend of decrease from high to low throughout the growing stage,but there was no differences isozyme bands of two ploidy Siraitia grosvenorii.Diploid E2and E4in24-50days still played more improtant role in promoting the enlargement of the fruits.3The fruits of diploid and triploid Siraitia grosvenorii were regulated by using exogenous hormones of IAA,GA3,ABA and ZR. The results showed that the most significant experimental factors which affected the fruit weight and the sweet glycosides V of diploid and triploid Siraitia grosvenorii were spraying intervals and spraying time. In terms of fruit weight for the assessment indicators in production, the best experimental treatment combinations to promote the diploid weight was the ABA (150mg/L, starting from first vine was cut off the top,spraying1time in total) that made the weight of the fruit significantly increase by21.18g.But for the triploid fruit the the best experimental treatment combination was GA3(25mg/L of GA3, starting from the third ceman female flowers were pollinated,spraying1time in every5days,2times in total) which made the weight increase by9.74g.In terms of sweet glycosides V for the assessment indicators in production, the most significant experimental treatment combinations wasZR(150mg/L, starting from the second ceman female flowers were pollinated,spraying1time in every5days,spraying2time in total) that made sweet glycosides v respectively increased by22.65%and29.29%.4The homologous gene of LEAFY of Siraitia grosvenorii were studied in this paper. Tow fragment named Sgl-LFY and Sg2-LFY gene was obtained. The Sgl-LFY of DNA clone contained a complete sequence of973bp LEAFY homologous gene which included2exons (369bp and340bp)and1introns(264bp) and encoded236amino acids. The Sg2-LFY of DNA clone contained a complete sequence of729bp LEAFY homologous gene which included1exons (310bp)and1introns(413bp) and encoded101amino acids.Sequence comparisons by performing Blast Search in GeneBank revealed that the two DNA gene has a high identify of77%to97%with LFY homologous gene of other plants.After the deduced amino acid sequence of the exons were submitted to Blast Search in GeneBank with LFY homologous gene of other twenty plants, it was found that the homology reached73%to97%. It was showed that the fragment gene belonged to LEAFY homologous of Siraitia grosvenorii indeed.And LFY homologous genes of different plant were conservative, also differences.
