Study On Genetic Diversity And Chalcone Isomerase Gene Diversity In Tartary Buckwheat Germplasm Resources

Abstract/Summary:
Tartary buckwheat has a long history of cultivation in China, and is the only crop as both food and medicine. There are rich resources of tartary buckwheat in southwest China of Sichuan, Yunnan, Guizhou and Tibet. However, we have not collected and saved tartary buckwheat resources enough and developed tartary buckwheat resources completely. So we need to do a lot of research about tartary buckwheat to fill the vacancy.In this study, we developed and designed500pairs of primers, and tested in tartary buckwheat. The effectiveness of these SSR primers in tartary buckwheat is about50%, the polymorphic rate is10.8%and the average value of PIC is0.4537. We analyzed the genetic diversity of core collection of tartary buckwheat with28SSR markers. A total of85alleles were identified, ranging from2-5with averaged3per locus. Shannon’s information indices of geographical variation were0.3633-0.6671. The accessions from Yunnan, Sichuan and Tibet are not only abundant in genetic diversity but also have the close phylogenetic relationship. This further confirmed that tartary buckwheat originate in the Southwest of China. This study demonstrates the method of development of SSR primers in tartary buckwheat would be very effective, and will contribute to the researches on genetic diversity analysis, identification of useful traits and marker-assistant breeding in tartary buckwheat.Tartary buckwheat has become a popular health food in China. Tartary buckwheat is rich in flavonoids and had a significant effect on a variety of diseases. However, Chalcone isomerase is one of the key enzyme in flavonoids biosynthesis of Tartary Buckwheat, but still not be researched enough now. In this study, according to the NCBI we know part of the sequence of chalcone isomerase of tartary buckwheat. And we designed3pairs of specific primers to amplify the full-length sequence of chalcone isomerase by chromosome walking technology. We designed another primers and analyzed gene diversity of chalcone isomerase in35accessions of tartary buckwheat. Chalcone isomerase gene sequence was amplified to1610bp, then we checked all the chalcone isomerase gene sequences using software, and analyzed the struncture. We found there are4exons and3introns, which is typical struncture. We aligned35accessions of tartary buckwheat, analyzed the sequence diversity and found7SNPs,13In/del, which revealed chalcone isomerase gene sequence is more conservative. Further, analysis of linkage disequilibrium, P>0.1, indicating no significant differences. All suggested variation is less in CHI gene.
