Elymus sibiricus L. belongs to Genus Elymus, Gramineae family and plays an important role in thehusbandly and ecological environment protection. However, irreversible changes of seed occur to agingin conservation process because of the comprehensive function under the seed itself and outsideenvironmental factor. Seed aging will not only influence the seed physiological and biochemicalcharacteristics changes, but also influence integrity of seed genome DNA. Seed vigor, physiological andthe integrity of genomic DNA were discussed after artificial aging in the (58±1)℃hot water. Theresults of experiment were as follows:1. The optimal time to be appropriate for artificial aging of Elymus sibiricus L.seed was3~6minin the (58±1)℃hot water. The time is the inflection point to cause each seed vigor index changes.With the increase of aging time, vigor indexes present the declined trend. The decline of the relativegermination potential was other vigor index and extents declined were greater than other index,followed by relative germination ratio index, relative vigor index and relative germination rate. With theextension of aging times, the plantule and radicle length became shorter. Effect on plantule length hasgreater degree of influence than radicle length caused by seed aging. Seed seedling growth could beused as the one of indexes to evaluation seed vigor.2. In (58±1)℃hot water after artificial aging, conductivity of seed and vigor index were positivecorrelationship. Soluble sugar content and vigor index were not correlationship. MDA content and thevigor index had negative correlationship and the correlationship was significant or extremely significantdifference. Conductivity and soluble sugar content of seed could be used as evaluation index of Elymussibiricus L. or not, which needs to be studied further. MDA content can be used as evaluation vigorindex of Elymus sibiricus L. seed.3. Dry seed genomic DNA was extracted after artificial aging to be used for ISSR-PCR geneticanalysis of Elymus sibiricus L. germplasm resources. The changes of genomic DNA fragments in dryseed of Elymus sibiricus L. caused by seed aging mainly concentrated in750bp~2000bp and250bp~500bp and showed that macromolecular fragments of DNA disappeared and color became shallowgradually. And DNA fragments lost and increase were random caused by seed aging.4. The genetic parameters of four different germination rates populations of3accessions Elymussibiricus L. germplasm were tested by8ISSR primers. The genetic parameters including polymorphicpercentage, numbers of alleles per loci, effective numbers of alleles per loci, genetic diversity index andShannon’s index declined with germination rate dropped. Contrasted with CK, numbers of alleles perloci and part of genetic diversity index were significant different when germination rate dropped to10%.The rest of each parameter was not obvious or extremely significant difference. Low germination ratewas not conducive to reproductive update of Elymus sibiricus L.seed.
