In this experiment, we take Research the Genetic integrity of Elymus sibiricus L.in the aging method of 100%RH high temperature and high humidity (AA) to treatment 4 of Elymus sibiricus L.which in different gradient, respectively from the agronomic traits, physiological and biochemical indexes and genetic integrity of Elymus sibiricus L. seed aging.1. The vigor index of Elymus sibiricus L.had a degreesive tendency with prolonging of the aging to deal with Elymus sibiricus L. The soluble suger content of Elymus sibiricus L. had been increased after decreased in a short period of time as the aging gradient extension. The MDA increased with the longer aging. The study showed that with exacerbation of artificial aging, POD activity had been no rule change, SOD activity had been increased and then reduced in the later stage, and CAT activity was weaken after aging.2 After aging treatment on the seeds of transplanting seedlings, in the second year after reviving in addition to El-5 internodes looks for the control E1-0 significant difference, El-3 rachis node number compared with the control El-0 significant difference, others phenotypic differences not significant.3 This experiment,20 primer SSR used to analysis showed that,4 samples of Elymus sibiricus were detected in 277 loci, of which there are 229 polymorphic loci, alleles accounted for 82.67%; the average Shannon’s index was 0.5523, Nei’s gene diversity index average variation is 0.3396,4 copies of Elymus sibiricus with abundant genetic diversity; aging sibiricus seed compared with the control, with the deepening of the degree of aging, the aging of the gradient of the amplified loci, polymorphic loci and the percentage of polymorphic loci were decreased, genetic diversity reduced aging sibiricus seed; the genetic diversity parameters of the aging population, the number of alleles per locus, each point of the effective number of alleles, genetic diversity index and Shannon index including decreased, and the gradient is proportional to the increase of aging, change sibiricus seed genetic structure occurred after aging, reduce the frequency of genetic variation, genetic integrity is destroyed4 This experiment,8 primer SSR used to analysis showed that:4 sibiricus seed detection alleles for 571 polymorphic loci,501 alleles, accounting for 87.74%, Nei’s gene diversity index average variation was 0.2642, the average Shannon’s index was 0.4104,4 sibiricus seed have abundant genetic diversity; compared with the control group, aging sibiricus seed amplified loci, polymorphic loci and the percentage of polymorphic loci reduced, less genetic diversity; genetic diversity parameters analysis showed that, the aging sibiricus seed number of alleles per locus, each the effective number of alleles, genetic diversity index and Shannon index decreased, aging degree is deeper, more decrease, change sibiricus seed genetic structure occurred after aging, reduce the frequency of genetic variation, genetic integrity is destroyed.
