In this study, a self-crossing populations which include92strains was constructedby the single spores which was obtain from Flammulina velutipes1123, a back-crossingpopulations which include14strains was constructed by the same single spores withW23which was obtain from Flammulina velutipes8801.During the fruiting test,81strains get fruiting bodies include69self-crossing strainsand12back-crossing strains. After fruiting test, collected the data of agronomic traits andlysine content for genetic analysis.Compared whit1123,higher mycelium growth rate andfruiting bodise lysine contnet,smaller cap diameter was founded both in self-crossing andback-crossing populations.Other agronomic traits was worse than1123,except productioncycle.In the self-crossing, the production cycle was as the same as1123,but the productioncycle of back-crossing was longer. With correlation analysis,some low positive correlationbetween production cycle and fruiting body lysine content,stipe length and stipediameter;low negative correlation between stipe length and production cycle, yield andproduction cycle, yield and fruiting body lysine content was founded.No correlation wasfounded between the other traits.The coefficient of variation analysis found that thecoefficient of variation of Flammulina velutipes mycelium growth rate is large and thecoefficient of variation of production cycle is small.The variation in self-crossing andback-crossing was similar,but white-offsprings have more variation thanyellow-offsprings.By the gray relational degree analysis of the tow populations, identified53/81andW105as ideal strains, which similar to8801on agronomic traits but have higher lysinecontent.51/54,9/49was identified as ideal strains because of their agronomic traits aresimilar to FL.19.However, the fruiting period have more than20days in advance. Inaddition to these4strains,another3strains which have higher mycelial growth rate,2strains which have longer stipe length,2strains which have thinner stipe diameter,2strains which have thinner cap,2strains which have higher yeild,5strains which havehigher lysine content were geted.12winter mushrooms were collected as test samples from12different farms.Fruiting body lysine content were tested by two method, one is adopt automatic aminoacid analyzer, the other is use ninhydrin. With correlation analysis between the two methods’s results, correlation coefficient value is0.979, which means both the twomethods’s is highly positively correlated. Because of the high cost by use automaticamino acid analyzer, use ninhydrin detect lysine content is suitable for large-scalescreening.
