Liriope Muscari (Decne.)Bailey, which belongs to Liriope genus of Liliaceous, make medicinal andornamental in one set. Its earthnut is source of saponins, polysaccharides and other ingredients; it could beused as a foliage plant and is an important ground cover plant. The aim of this experiment is to takevarieties of Liriope Muscari (Decne.)Bailey as explants, in previous studies, discusses the mechanism ofseeding induction, callus culture, subculture proliferation, cultivation and rooting culture. Studying thechange of content of endogenous hormone when add exogenous hormone (6-BA, NAA,2,4-D).Throughstudying the mechanism of organizational culture, the rate of bud induction can be effectively improved. Itis good to form a series of Liriope tissue culture system, in order to carry out the industrializedproduction.The main research results are as follows:(1)Best disinfectant methods in Liriope Muscari (Decne.)Bailey is soaking in75%alcohol for30s,0.1%Hgcl2disinfection of8-16minutes. Sprouting seedling explants in Seed Germination Seedlingpollution rate, the highest survival rate. Liriope explant collection to consider seasonal, stem tip bestcollection season is March.(2)The best stem tip bud induction medium was MS+6-BA1.0mg·L-1+NAA0.2mg·L-1, induce thehighest rate, sprouting time is rapider,6-BA induced a significant effect.The content of ZT is reduced gradually, the content of GA3first increased slightly, and then graduallydecreased, has been kept at a low level, low concentration GA3growth beneficial to the buds. The contentof ABA has been at a lower level of hormones. IAA/ZT, IAA/GA3, ABA/GA3at a relatively stable level,IAA/ABA at a high percentage and has been increasing.(3)The best subculture multiplication formula is the MS+6-BA4.0mg·L-1+NAA0.2mg·L-1,proliferating coefficient reaches6.09.6-BA is the influence of Liriope increment coefficient factors.Maximum multiplication factor reach to the mix value when the subculture algebra in the sixthgeneration.In the progress of proliferation culture, the change of ZT hormone content first tall and late low. TheGA3hormone levels change is relatively complex, after rising to decline. IAA hormone content firstdecreased and then increased, but the change of hormone content not big. ABA hormone levels arerelatively stable, content has been increasing.(4)The best seedling formula is the MS+GA30.01mg·L-1+6-BA0.01mg·L-1, with the concentration ofhormone combination seedling effect is GA3+6-BA> GA3+IBA> GA3+NAA, high concentrations ofGA3inhibit seedling growth.The content of ZT decreased at first and then gradually smooth. The content of GA3increasedsignificantly. The content of IAA changed little early stationary, then fall. IAA, ABA smooth curve, thehormone content has been in a relatively low level.(5) The best rooting formulation is1/2MS+IBA1mg·L-1+sucrose30g·L-1, rooting rate was60.42%,the average root number up to8.13.The kinds of culture medium, IBA, sucrose are all have a significanteffect on rooting rate, the rooting rate of the primary and secondary relationships were medium> sucrose> IBA> NAA.Ac powder on rooting rate had no significant effect, but can promote Tissue Culture Plantlet Rootingin advance.Rooting effect of ABT1is better than ABT3in rooting powder,the ABT1concentration of2mg·L-1havethe best rooting effect, rooting rate is67.45%, in the1-3mg·L-1range, with rooting powder concentrationincreased, Liriope muscari rooting rate increased first and then decreased, rooting powder at low levels, theroot is weak, poor seedling, roo ting powder with high content and inhibit seedling growth.(6) Effect of different transplanting substrate formula on Liriope muscari transplanting survival ratehas a significant effect, the survival rate of transplanting in red soil: perlite: sand=6:3:1matrix is thehighest, up to93.33%.(7)Callus induction in the best match of MS+2,4-D1mg·L-1+6-BA1.5mg·L-1+NAA0.5mg·L-1+sucrose20g·L-1.2,4-D,6-BA, NAA are on the callus induction rate influence. Effects of primary andsecondary relations is sucrose>2,4-D>6-BA> NAA. Different explants have significant effect on callusinduction; the best material for inducing callus is stem tip from Radix Liriope, induction of100%.In the progress of callus culture, with the proportion of the IAA and ZT moderate can help callusformation. ABA content should be less and relative stability, lower concentration ABA in favor of callusformation, high concentration is not conducive to the formation of callus.(8)The best treatment of callus subculture is cultured on the MS+2,4-D1mg·L-1+6-BA1mg·L-1+NAA1mg·L-1growth is0.98g. The result of range analysis is R2,4-D>R6-BA>RNAA.2,4-D、6-BA、NAA have the primary and secondary influence on the callus induction rate perform of sucrose>2,4-D>6-BA> NAA.During callus culture, IAA and ZT as the proportion of moderate contribute to callus formation. ABAcontent is relatively stable and less content, ABA content is too high and not conducive to callus formation.Low concentration in favor of callus formation(9) The best effect of Callus budding is the treatment of MS+6-BA4mg·L-1+NAA0.2mg·L-1, and havea similar result induced by shoot tip proliferation culture.6-BA play a very significant role on callus budinduction.
