Quercus variabilis has a widespread distribution area in temperate and subtropical areas in China. It is the most important cork resources species and important ecological and economic values. In this paper,5-year-old plus trees open-pollinated progenies from five Q. variabilis natural populations as materials, phenotypic traits differences were invesitigated for different populations and families. With analyzing peroxidase isozyme, the variations of the nitrate reductase, soluble protein, chlorophyll as well as soluble sugar were determinated. In order to explore the future seed source for early selection and shorten the breeding cycle, it provided a theoretical basis. The main results are as follows:(1) There were significant differences in the nitrate reductase activating, the content of soluble protein, the chlorophyll content and the content of soluble sugar among the different populations (P<0.05). Meanwhile, the same provenance varied with the seasons obviously. With higher values of these four indices, HeNan BaoTianman provenance is considered to be a good provenance.(2) In July, several physiological indexes of families reached the maximum, which was significant (P<0.05). There was great positive correlation between sapling tree height and the nitrate reductase activating, the content of soluble protein, the chlorophyll content and the content of soluble sugar, and correlation coefficients were0.547、0.643、0.492, which could be used as physiological indices in early selection of Q. variabilis.(3) With principal component analysis and cluster analysis of Q. variabilis, Shaanxi Ninying and Shaanxi Taibai came together and were more recent genetic distance to Henan Xinyang. And they came together with Shaanxi Lou Guantai. But Henan Bao Tianman was relatively largest with four other group of genetic distance.(4) Through cluster analysis in July, the family of Q. variabilis can be divided into five major categories. From the physiological and biochemical indexes, Henan Baotianman family distinguished with other familes and each class was basically the same source. It provided a theoretical basis for choice of excellent pedigree.
