This test studied the optizimation of the embryoids culture condition via in vitro cultureof unpollinated ovary and plantlets formation for5genotypes of summer squashmaterial.Mainly discussed the influence of the embryoid from genotype、medium type、hormone kind and concentration,and optimized the culture conditions.Take morphologicobservation、root striked and domestication for the embryo sac regeneration plant tubeseedlings which were abtained from the test.The results showed that:1. Summer squash genotype has a certain effect to embryoid induction rate and embryoidinduction time.2.Induced effect of in vitro culture of unpollinated ovary was the best in the MS medium.3.Hormone was necessary to the in vitro culture of unpollinated ovary.It could not induceembryoids with hormone-free medium. It was the best to add1mg/L6-BA+0.5mg/LNAA+1mg/L2,4-D.4. The concentration of NAA affected root quality and length,adding0.1mg/L NAA wasbetter,the root can grow strongly and only need11days can seen visible roots,reached7-10cm.5.Plantlets with3-6true leaves,first half-open bottle ang then completely open the bottletwo days later,moved to the nutrition bowl buried in the domestication of7days in greenhouse soil before planting,the survival rate reachered more than95%.6. Regeneration plants’plant morphology showed,haploid plants grow slowly,and the sizeof organ is smaller,male flowers have no pollens,stigmas are deformed; Double haploid plantsand heterozygous diploid plants were growing normal,double haploid offspring plants wereno separation;Aneuploid plants showed sterility;Tetraploid plants exhibited significant.
