Tobacco virus disease is one of the very important diseases in tobacco planting areas all over the world, which could have serious consequences on the yield and quality of tobacco. Hubei province gives high yield of tobacco, and the incidence of tobacco virus disease has been more serious in recent years. To make certain the species of tobacco virus disease in Hubei, some of varieties of tobacco virus species on366collected samples which infected tobacco in Shiyan, Xiangyang, Enshi, and Yichang planting areas have been identified indoor on the base of the field investigation.1. Mosaic, distortion, lightning stripe, vein necrosis, etching, and ring spot are the main kinds of tobacco virus diseases symptom, in Hubei province, and mosaic disease took the first place, and the others showed a lack of incidence, in addition mixed pattern with two or more kinds. The mosaic disease was the dominant in Shiyan and Xiangyang. Both the mosaic and the distortion disease were the dominant in Enshi and Yichang. The ring spot were found in Xiangyang only at the present time.2. The result of indication plants idenfication showed that there were kinds of symptom displayed on the10species of host plants virus, including mosaic, vein clearing, distortion, local necrosis, chlorosis, etching, and ring spot. Moreover, different types of virus disease symptoms displayed discrepancy on different plants, and it could be concluded that the samples contained Tobacco mosaic virus (TMV), Cucumber mosaic virus (CMV), Potato X virus (PVX), Potato Y virus (PVY), Tobacco etch virus (TEV), and Tobacco ringspot virus (TRSV).3. The result of serology demonstrated that the detection rates of TMV, CMV, PVX, PVY, TEV, TRSV were92.1%,42.1%,45.6%,35.0%,22.4%,12.6%, respectively, and the dominant TMV had the highest rates. The distribution of the virus and the superior among all the collected areas differed a little. The rate of mix-infection was70.8%, and the patterns could be classified into29types, and TMV+PVX took the first place. The rate of mix-infection and the superior among all the collected areas differed a lot.4. The situation of virus disease was not the same on different speices of tobacco. The rates of TMV, CMV, PVX, PVY, TEV, TRSV detected from cured tobacco samples were85.5%/47.1%,51.6%,24.0%,22.2%,19.9%, respectively. The detection rate of mix-infection was65.6%, and the patterns could be classified into25types, and TMV+CMV+PVX+PVY+TEV+TRSV took the first place. The rates of TMV, CMV, PVX, PVY, TEV, TRSV detected from burley tobacco samples were93.2%,37.5%,36.4%,23.9%,21.6%,2.2%, respectively. The rate of mix-infection was65.9%, and the patterns could be classified into15types, and TMV+PVX took the first place. The rate of TMV, CMV, PVX, PVY, TEV detected from57Maryland tobacco samples were100%、29.8%,36.8%,94.7%,21.1%, respectively. The rate of mix-infection was98.2%, and the patterns could be classified into7types, and TMV+PVY took the first place.5. The result of molecular biology showed that TMV samples with313bp got98.4%~100%nucleotide sequence identity and96.81%~99.68%compared with registered TMV CP gene sequences. CMV samples with326bp got93.6%~100%nucleotide sequence identity and92.64%-97.55%compared with registered CMV CP gene sequences. PVY samples with825bp got98.9%~100%nucleotide sequence identity96.85%~97.45%compared with registered PVY CP gene sequences.The quarantine inspections in nation, TRSV and in province, TEV were firstly declared in the research, so as the field investigation to the virus disease of Maryland tobacco in China. All above were hoped to provide some definite theoretic basis for the management and control of the tobacco virus disease in Hubei province.
