| BACKGROUND of MH63::15:Brown planthopper (Nilaparvata lugens Stal; BPH) resistance genes Bph15have been introgressed singly or pyramided into rice variety Minghui63(MH63).This article studied the influence of MH63::15to these four genes (exuã€magã€vasã€vg), in order to know the changes of these genes relative expression from molecular level.In this paper we used Minghui63(MH63) as the control, which was the susceptible. First, BPH was fed on MH63and MH63::15.Then the ovaries were dissected and classified. At last, the RNA of ovaries were extracted. The results of real-time PCR showed the changes of the four genes relative expression. The results were as follows:(1) The results of females’ovarians classification showed that compared with the BPH fed on MH63, the ovarian growth speed of whice were fed on MH63::15was slow. Moreover, the proportion of the egg-laying stage (Grade â…£) decreased significantly.(2) We detected the changes of the four genes related to oogenesis expression through real-time PCR. The results were as follows:exu:The expression of BPH fed on MH63::15was much higher than that fed on MH63in females. MH63::15was a opposite regulator of exu gene expression.mag:The expression of BPH fed on MH63::15was much lower than that fed on MH63in females. MH63::15was a negative regulator of mag gene expression.vas:The expression of BPH fed on MH63::15was much lower than that fed on MH63in females. MH63::15was a negative regulator of vas gene expression.vg:The expression of BPH fed on MH63::15was much lower than that fed on MH63in females. MH63::15was a negative regulator of vg gene expression.The result of expression of genes showed that except exu, the other genes’expression of BPH fed on MH63::15was reduced, the synthesis of protein related to ovary development was reduced, So the development of ovaries were slowed down. |
