Rhododendron Chrysanthum Pall, have ornamental and economic value that belongs to the Rhododendron hybrides. Its roots,stems and leaves contain tannin and aromatic oil. It can refine quebracho extract and can made into tea. So that Rhododendron Chrysanthum Pall, have very high research value. This experiment use tissue culture, anatomy and genetic diversity of three methods. The results are as follows:1. Through tissue culture method comparison of seed emergence rate of Rhododendron Chrysanthum Pall. in Changbai Mountain and Laobai Mountain. Known that Changbai Mountain’s seed emergence rate was higher than Laobai Mountain’s. Germination time of tissue culture seedling stems earlier than leaves and stem’s callus induction and differentiation rate also higher than leaf’s. So the bests material for callus induction was stem. Rhododendron Chrysanthum Pall. transplanting seedling survival rate was relatively low that life was poor and the plant grows slow and leaves variable in yellow. Under different irrigation method known that given a certain amount of nutrition liquid was good to transplanting seedlings.2. Analysis on different altitude in Changbai Mountain of Rhododendron chrysanthum Pall. leaf anatomical structure under the microscope show that leaf thickness,palisade tissue thickness,thickness ratio of palisade to spongy tissue,Cell tense ratio of leaf and Stomata density with increasing altitude showed a trend of increase while the spongy tissue thickness and Spongy ratio of leaf has a contrary result.3. Research on Rhododendron chrysanthum Pall. genetic diversity with RAPD technology show that12primers in11tested groups amplified136strips that131bands were polymorphic bands that accounted for96.32percent of the total. The26individuals on elevation of1900m in Changbai Mountain were amplified97bands that88bands were polymorphic bands that accounted for90.72percent of the total. The23individuals on elevation of2370m group were amplified123bands that107 bands were polymorphic bands that accounted for86.99percent of the total. The25individuals on elevation of2530m group were amplified112bands that98bands were polymorphic bands that accounted for87.5percent of the total. On elevation of1900m,2370m and2530m of three groups that74bands individuals were amplified152bands that151bands were polymorphic bands, accounted for99.34percent of the total.The population between11groups of tested materials genetic similarity coefficient range is0.4559to0.7574. The elevation of1900m,2370m and2530m in Changbai Mountain internal group’s genetic similarity coefficient of variation were0.5773to0.8454,0.5447to0.8374and0.5536to0.8571.Cluster analysis results show that at the similarity coefficient of0.61where the tested of10groups can be divided into two groups the first for Changbai Mountain and second for Laobai Mountain.Three populations of74individuals in Changbai Mountain comprehensive analysis show that population generally grouped into own categories. Nine populations of84individuals in Changbai Mountain comprehensive analysis show that at the similarity coefficient of0.62can be divided into three groups. The first category is the elevation of1900m,the second category is the elevation of2370m and the third category is the elevation of2530m.A,B,C and D are gathered for the first group. D+,E,F and G are gathered for the second group. H and I are gathered for the third group. Rhododendron chrysanthum Pall, individual of similar elevation has smaller genetic distance and larger coefficient of genetic similarity.
