The emergence of crown buds in herbaceous peony is important for the propagation.Using plants through division, rooted cuttings and seedlings of herbaceous peony, this studymainly evaluated the effects of different technique on the crown bud emergence of herbaceouspeony cultivars. The main contents and results were as follows:1. For the plants through division, we mainly used two techniques separately, dividingplants and treating with6-BA solution. The cultivars used were ‘Da Fu Gui’、‘Qi Hua LuShuang’ and ‘Yang Fei Chu Yu’.To study the effect of division on the bud emergence, five or six–year were used. Theresults mainly showed that with plant-dividing treatment:(1) Bud number increased. Theratio of bud number to stem number of the three cultivars were1.25、1.27and1.40respectively.(2) Germination rate of ‘Da Fu Gui’ and ‘Yang Fei Chu Yu’ decreased whereasthat of ‘Qi Hua Lu Shuang’ increased. To examine the effect of6-BA treatment on the bud emergence, three concentration of6-BA,300mg/L-1、600mg/L-1、900mg/L-1, was respectively used by watering the testedcrowns. The result showed that:(1)6-BA promoted bud emergence and the optimalconcentration varied with cultivars. For ‘Da Fu Gui’300mg/L-16-BA produced the besteffect by increasing0.18more buds; For ‘Qi Hua Lu Shuang’600mg/L-16-BA wasoptimum by increasing1.18more buds; For ‘Yang Fei Chu Yu’900mg/L-16-BA wasoptimum by increasing0.35more buds.(2)6-BA treatment advanced bud sprouting by0-3d.(3) the induced buds by6-BA treatment mainly came from both the mother bud and thegrandmother bud.2、The rooted cuttings varieties were ‘Da Fu Gui’、‘Qi Hua Lu Shuang’、‘Yang FeiChu Yu’. Experimental content including the following several aspects:(1) Study on theinfluence of different sites on the germination by cut the stem into three section.(2) Thecutting seedlings were irrigated root by different concentrations6-BA in the field andgreenhouse conditions, study on the difference of rooted cuttings germination between thefield and greenhouse conditions. The results showed:(1) Bud occurred in leaf axils located the cuttings base When theupper part of the stem segment as cuttings randomly. The analysis thinks that cutting leafstimulate dormant bud in leaf axils to sprout.(2) There was no bud occurred on roots of thecutting seedlings which treated with6-BA in the field and greenhouse conditions. This may bedue to the relatively late time when rooted cuttings were moved to the pot, thus affectedadventitious bud germination. In addition, we used ordinary garden soil which lacked airpermeability as matrix, this may be one of the reasons for failure.3、Seedlings which were one year old and two years old came from the seedling of thespecies ‘Zi Feng Yu’ as the materials. Experimental content including the following severalaspects:(1) influence on the seedlings underground buds after irrigating root by differentconcentrations6-BA.(2) Influence on buds occur after removing biennial seedling apical bud.(3) the change of soluble protein and soluble sugar in buds and roots during buddifferentiation. The results showed:(1)6-BA had a distinct influence on the seedlings. The buds andstems number increased, the height decreased in within the treatment concentration.According to analysis, because6-BA breaks the inhibition of the top buds to the lateralinhibition, thus the lateral bud grew. Simultaneity, the parts of on the ground and undergroundlose balance, in turn the height was reduced.(2) Removing biennial seedling apical budpromoted lower buds occurrence,1.2more than contrast.(3) Soluble protein and solublesugar in buds and roots during bud differentiation first increased and then decreased, according to the analysis, variation trend had a connection with material needs. In productionpractice, supplement adequate nutrients in time can promote plump buds and plants haleness. In conclusion, we mainly examined the impact of different techniques upon the crownbud emergence in the different sources of peony plants, meanwhile we determined nutrientcontent during the whole stages of crown bud differentiation. The research results willcertainly provide practical guide for promoting crown bud emergence in herbaceous peony.
