Underground Buds Induction And Callus Cultivation Of Paeonia Lactiflora Cultivars

Paeonia lactiflora has high ornamental and officinal values. In this experiment, five excellent herbaceous peony cultivars,'Tuan Ye Hong','Zhong Sheng Fen','Fen Yu Nu','Da Fu Gui' and 'Zhu Sha Pan', were chosen as experimental materials to study initiation culture of underground buds, multiple shoots induction and proliferation, rejuvenation of hyperhydric shoots, improving the growth of shoots in summer, rooting, transplanting, callus induction and differentiation. Main results were as follows:(1)'Zhu Sha Pan' behaved the best among the five herbaceous peony cultivars in initiation culture. IAA was in favor of initiation culture.1/2 MS (double-strength Ca2+)+6-BA 1.0 mg/L+GA3 0.5 mg/L +IAA 0.2 mg/L was found to be the best initiation medium for 'Zhu Sha Pan'; 1/2 MS (double-strength Ca2+)+6-BA 1.0 mg/L+GA3 0.5 mg/L+IAA 0.1 mg/L was the best medium for 'Zhong Sheng Fen'. It's better to sample explants in spring than in winter. Low concentration of 6-BA (0.5 mg/L) should be used in initiation culture of 'Da Fu Gui'.(2) Resection flower bud and leaves was favorable for multiple shoots induction of 'Zhong Sheng Fen'. The best multiple shoots induction medium for 'Da Fu Gui' was 6-BA 1.0 mg/L+KT 0.5 mg/L. 6-BA 1.0 mg/L+KT 0.5 mg/L+IAA 0.5 mg/L was the best medium for proliferation of 'Zhu Sha Pan'. 0.2 mg/L 6-BA was the best treatment for 'Zhong Sheng Fen' in multiple shoots proliferation.(3) Some methods allowed hyperhydric plantlets of 'Zhong Sheng Fen' to rejuvenate and develop normally, such as the addition of 3 g/L activated charcoal, removal of ammonium nitrate from the medium, doubling the concentration of Ca2+, eliminating BA from the medium and using joint filling material. Double concentration of Fe2+ was a good method for overcoming yellowing of the shoots.1/2 MS (double-strength Ca2+) or WPM medium without any hormones were favorable for improving the growth of the weak shoots in summer.(4)The best medium for rooting of 'Zhu Sha Pan'and' Da Fu Gui'was 1/2 MS (double-strength Ca2+)+IBA1mg/L.(5) The best callus induction medium of'Da Fu Gui'was 6-BA 1 mg/L+NAA 0.5 mg/L+2,4-D 0.5 mg/L.6-BA 1.0 mg/L+NAA 1.0 mg/L and TDZ 0.5 mg/L+NAA 0.5 mg/L were the best treatment for callus induction of'Fen Yu Nu'. The best medium for callus proliferation was TDZ 0.5 mg/L+2, 4-D 0.5 mg/L. Adding 100 mg/L CH or LH was good to the development of callus. Liquid culture was better than solid culture and WPM medium was for callus proliferation and growth.2,4-D inhibited differentiation.
