Chinese Cabbage (Brassica campestris L. ssp. pekinensis Lour) belongs to cruciferaebrassica and it’s one of the most important vegetables origining in China, TuMV(turnipmosaic virus) is one of the most serious diseases harm to chinese cabbage in cultivation. Inthis study, the inheritance, physiology nad molecular mechanisms of Chinese cabbageresistance to TuMV were studied, the main results were summarized as follows:1.Two resistant(antigenic material8407and highly resistant material73) and twosusceptible(Guan291and Chunyuehuang) materials were used to make4F1hybrids,then theF2segregations were constructed from the4F1hybrids.Above-mentioned plants wereinoculated with TuMV-C4strain.TuMV resistance of Chinese cabbage were tested by usingthe method of visual observation,condition level and classification standard.The resultsshowed that,the F2segregations have different resistance to TuMV,which derived from the F1plants between the same resistant material and two different susceptible parents.In thecombination of8407×Guan291,TuMV resistance was controlled by a single dominantgene.In the combination of8407×Chunyuehuang,TuMV resistance was controlled by a pairof recessive genes.In the combination of73×Chunyuehuang,TuMV resistance wascontrolled by a pair of recessive genes.In the combination of73×Guan291,TuMV resistancewas controlled by tow pairs of recessive genes.2.The4materials (8407,He304,Guan291and Chunyuehuang) which have differentresistance to TuMV,were chosen to be the material plants.They were inoculated withTuMV-C4strain at seedling stage.The4materials’ enzyme activities of SOD、POD、CAT andthe content of H2O2were detected within24days after inoculated by TuMV.The resultsshowed that TuMV resistance of Chinese cabbage had the most closely relationship withH2O2and CAT,and had less relationship with POD,and had no relationship withSOD.Therefore,detection of the change of the content of H2O2and enzyme activities of CATand POD is valuable to the initial screening of TuMV resistance of Chinese cabbage on someextent.3.Plant materials were the same as those used in the second section. The real time quantitative RT-PCR of CAT1~3genes in Chinese cabbage were performed. The resultsshowed that the CAT2and CAT3genes may be play an important role in the total enzymeactivity of CAT in seedling stage leaves.In this study,the result of the real time quantitativeRT-PCR of CAT genes showed almost the same trend with the result of enzyme activity ofCAT, suggesting CAT played an important role in the proceeding of resist to Virus, whenChinese cabbage was infected by TuMV.
