Association Of Pcr-Rflp Polymorphisms Of MC1R Gene With Plumage Colour In Lingshan Local Chicken

The Lingshan chicken was investigated using an intercross between the the Lingshan chicken and the Wens yellow chickens. The homozygous Lingshan chicken F1generation only have blackpock feather while yellow chickens were found in the heterozygous Lingshan chicken F1generation four plumage colors (black, black with yellow neck, blackpock and yellow) were generated after the F1generation blackpock chickens self-crossing. PCR-RFLP technology was applied to analyze the polymorphisms of MCIR gene in three-generation pedigree (parental generation:30homozygous Lingshan chickens,15heterozygous Lingshan chicken,30Wens yellow chickens; Fl generation:30blackpock chickens,15yellow chickens,3pale yellow chickens; F2generation:20black chickens,26black with yellow neck chickens,80blackpock chickens,20yellow chickens). The results provided compelling evidence that the Extended black (E) locus controlling plumage color is equivalent to MC1R. In the parental generation,The genotypes of30homozygous black-feather chickens were EE,.the genotypes of Lingshan heterozygous chickens were divided into two types:EE and Ee;30Wens401yellow chickens were ee; In the Fl generation, the genotypes of30blackpock chickens were Ee;15yellow chickens were ee, and3pale yellow chickens were Ee. We speculated that there were two other yellow genes BU and BU’which control feather Color except E. If the frequency of BU and BU’is higher than E, BU and BU’ will produce epistatic effect on the E, so the pale yellow chicken’s genotype may be Ee BUBUBU’BU’; the Lingshan heterozygous Chicken’s (Ee-type) genotype is Eebububu’bu’, the Lingshan heterozygous chicken’s (EE-type) genotype is EEBUbuBU’bu’. By studying the genotypes of F2generation, found that the chicken color was changed from black to yellow with the fall of E gene frequency. And the segregation ratio of offspring was basically according to genetic ratio.
