Research On Plumage Color Purification Method In Bashang Long-Tail Chicken Black Plumage Breeding Group And Application Model Of Barred Plymouth Character In Auto-sexing

Plumage color is an important economic trait.It plays an important role in determining the purity of given breed,hybridcombination and phylogenesis.Plumage color have not received much attention of breeders until recent years.Bashang Long-tail Chicken,one of Hebei Province local chicken breeds that display a rich plumage color types.However,the research and utilization on plumage color of Bashang long-tail chickens are very poor.In the paper,based on previous studies in our laboratory,both method of plumage color purification in Bashang Long-Tail Chicken Black Plumage breeding group and application model of Barred Plymouth character in auto-sexing were developed.The results will provide reference to the selection of new strain in Bashang Long-tail Chicken.1.Bashang Long-tail Chicken,Beijing Fatty Chicken and Leghorn Chicken were used as samples,and MC1 R gene was selected to amplify and sequence for different plumage color populations.Plumage color correlation,haplotype analysis and selection pressure analysis were performed on MC1 R gene mutation sites in different chicken populations.The results showed that the difference between 11 SNP site genotypes and plumage color were very significant or significant(P<0.01 or P<0.05),and the correlation between five haplotype combinations and plumage color was very significant(P<0.01).The Bashang Long-tail Chicken Black Plumage breeding group was subjected to positive selection pressure at three sites including c.274G>A,c.398T>A and c.427G>A.The analysis suggested that c.274G>A and c.427G>A may be important functional site of the black plumage trait.At the same time,a method to identify pure black individuals of Bashang Long-tail Chicken is established by combining black plumage trait linkage sites with TYR gene.2.The Bashang Long-tail chickens of Barred Plymouth hen and the non-Barred Plymouth cock were used as samples of study.The phenotypes of 1-day-old chicks were observed through cross combination and incubation,and CDKN2A/B gene of chicks and their parents were amplified and sequenced.It was found that Barred plymouth character of Bashang Long-tail Chicken conforms to the law of sex-linked inheritance,and two application models of auto-sexing of Barred Plymouth character of B1 W type and B2 W type were established.
