By the method of molecular biology,this work studied the bacterial community structure in the cecum of rhizomyidae,it will provide a fundamental theoretical on diversity and the isolation and purification of rhizomyidae cecal microorganisms and clone of functional genes.Total microbial DNA was directly extracted from contents of rhizomyidae cecal microorganisms. The clone library of16S rRNA genes was amplified using PCR with universal bacterial primer sets. The PCR products were then cloned into pGEM-T vector.Each unique restriction fragment polymorphism pattern,created by using two restriction endonucleases(Hha Ⅰ and HaeⅢ),was designated as an operational taxonomic unit(OTU).Amplified DNA was used for diversity analysis.Results showed that DNA extracted by freezing-thawing is excellent and find a rapid and accurate identification of positive clones.The distribution of rhizomyidae cecum microorganism is rich,Lactobacillus,Bacillus sp, fusiform bacillus and Paenibacillus polymyxa distribute in rhizomyidae cecal were contemporary.It also has some unknown and unculture strains.Unknown and unculture strain are dominant bacterials.
