Rabies is an acute viral disease to causes encephalomyelitis in virtually all the warm blooded animals including man, Once the onset of symptoms, there will be a high fatality rate of100%. The causative agent rabies virus is found in normal domestic dogs and cats, transmitted to other animals and/or to humans through close contacts with their saliva by biting, scratching, licking the injured skin and mucous membranes. Rabies occurs in all continents with the exception of Australia and Antarctica. Several countries are currently free of rabies, such as Japan, Australia, New Zealand, UK and so on, because the countries located in islands.In this study,325dog brain samples were collected from Guangxi and were used to detect rabies virus by RT-PCR. Totally10samples were diagnosed as rabies virus positive. Seven rabies virus were isolated from the positive samples: GXBH2011,GXLB2010,GXLQ2010,GXBS092010,GXBS132010,GXBS822010and GXBS892010by mouse injection test. The nucleotide sequences of Leader RNA to G-L interval region of six rabies virus isolates and complete genome sequence of GXBH2011were determined. The homology, the main antigenic sites, the B-cell epitopes, the Th cell epitopes, glycosylation sites and other sequence changes of the rabies virus were investigated, by comparing nucleotide and amino acid sequence with that of published rabies virus strains.By phylogenetic tree analysis, the results showed that the isolates GXBS132010, GXBS822010, GXLB, GXHXB and GXNND belong to group Ⅰ; the isolates GXBH2011, GXLB2010, GXLQ2010, GXBS092010, GXBS892010, GXBM, GXNN2and GX219belong to group Ⅱ. In this study, the complete genome sequence of rabies virus GXBH2011was amplified. The nucleotide sequence had high homology, when compared to FJ008, FJ009, SH06was more than96.1%, when compared to F02, GX4was more than87.7%. The GXBH2011was closely relative to the region of Fujian, Zhejiang in dogs with rabies virus by Phylogenetic tree analysis. At the same time, we analyzed the non-coding region of rabies virus GXBH2011, showing that rabies virus GXBH2011sequences are very conservative in the start and end sites, UUGU in the3’starting position, U7in the5’end position. The high conservative area is located at the3’and5’ends of the genome, and11terminal nucleotides of3’and51end of rabies virus genome match each other.
