Research On The Effect Of Exogenous ABA To The Cold-Resistence Proteins In Winter Wheat

Winter wheat is one of the major food crops in China, and notably affected by low temperature, thereby increasing the cold-resistance of winter wheat had become the focus of our research. In this study, we used strong chill-proof Dongnongdongmai1(reviving rate is higher than85%), which could live safely through winter under natural conditions, and weak chill-proof Jimai22(reviving rate is lower than2%) as experiment materials, exogenous ABA and natrium wolframicum treatments to the seedlings, and18℃,4℃,-6℃and-20℃low temperature stress. Determined the content of soluble protein in wheat tissues, and analyzed the total protein by polyacrylamide gel electrophoresis (SDS-PAGE). Investigated based proteomics of exogenous ABA on the impact of cold hardiness in winter wheat. The results as followed:(1)Effect of exogenous ABA to the content of soluble protein in two kinds of winter wheatsExogenous ABA could enhance the content of soluble protein in winter wheat. Exogenous ABA ccould significantly enhance the content of soluble protein in two kinds of winter wheat leaves, and the improving effect in the leaves of Dongnongdongmai1was better than in Jimai22. The content of soluble protein reached top (30mg/mL) by ABA treatment in the leaves of Dongnongdongmai1under-6℃. Exogenous ABA could notably enhance the content of soluble protein in the subterraneous stems of Dongnongdongmai1, and with temperature decreasing, it showed a gradual upward trend. The improving effect of exogenous ABA to the content of soluble protein in the subterraneous stems of Jimai22was weaker than in Dongnongdongmai1. The effect of exogenous ABA to the content of soluble protein in the roots of Dongnongdongmai1was not significant, but notably enhanced the content of soluble protein in the roots of Jimai22.(2) Exogenous ABA to the total protein in two kinds of winter wheats by SDS-PAGE analysisSpecific proteins with molecular weight of about23kD,35kD and51kD could be induced by exogenous ABA in the leaves of Dongnongdongmai1under low temperature, and proteins with molecular weight of about15kD,43kD and53kD were upregulated by ABA with the temperature decreasing. Specific proteins with molecular weight of about28kD and36kD could be induced by exogenous ABA in the leaves of Jimai22under low temperature, and proteins with molecular weight of about15kD,53kD and76kD were upregulated by ABA with the temperature decreasing. Under each temperature,18kinds of proteins were upregulated or specific expressed by exogenous ABA in the leaves of Dongnongdongmai1, comparing to Jimai22, and these proteins were the cold-related proteins, which were induced by exogenous ABA.Proteins with molecular weight of about19kD,20kD,21kD,24kD,38kD and44kD were upregulated by exogenous ABA in the subterraneous stems of Dongnongdongmai1under low temperature. Protein with molecular weight of62kD was specifically expressed in three treatments in Dongnongdongmai1, indicating that it was induced by low temperature and related to cold-resistance. Protein with molecular weight of about14kD was specifically expressed in three treatments in the subterraneous stems of Jimai22under low temperature. This protein was highly expressed in ABA treatment than the other treatments, and we can infer it was a cold-related protein, overexpressed by exogenous ABA. Proteins with molecular weight of about19kD and23kD were upregulated by exogenous ABA in the subterraneous stems of Jimai22under low temperature. Under each temperature,9kinds of proteins were upregulated or specific expressed by exogenous ABA in the subterraneous stems of Dongnongdongmai1, comparing to Jimai22, and these proteins were the cold-related proteins,which were induced by exogenous ABA.Proteins with molecular weight of about20kD and56kD were upregulated by exogenous ABA in the roots of Dongnongdongmai1under low temperature. Specific protein with molecular weight of about16kD could be induced by exogenous ABA in the roots of Dongnongdongmai1under low temperature. Cold-related proteins with molecular weight of19kD,20kD,21kD and78kD were expressed in three treatments in the roots of Jimai22at4℃, but with temperature decreasing, expressions of these four proteins were decreasing. Under each temperature,9kinds of proteins were upregulated or specific expressed by exogenous ABA in the roots of Dongnongdongmai1, comparing to Jimai22, and these proteins were the cold-related proteins, which were induced by exogenous ABA.
