Vegetables are the major source of nitrate intake. Scientists focused on minimizing the nitrate accumulation through agricultural management, screening genotypes, and post-harvest storage. Cucumber is one of the main vegetables in China, and to reduce the nitrate intake is an effective way.to to minimize the nitrate content of cucumbers. The results were followed:First, method of combination BSA with SRAP was used to find the molecular marker linked to the nitrate content of Cucumber. A F2population, derived from a cross between’1058’(higher nitrate content,190.71mg/kg·FW)and D0327(lower nitrate content,190.71mg/kg·FW) was used to analyze nitrate content through sulfosalicylic acid colorimetry, which followed normal disribution.Then, the genome DNA of cucumber was isolated through the improved CTAB method, which had a higher quality after the detection of1%agarose gel electrophoresis.A new stable cucumber SRAP-PCR reaction system was established by orthogonal design (15μL) including10×PCR Buffer1.5μL,25mM MgCl20.8μL, Taq polymerase (2.5U/μL)0.1μL, template DNA20ng, primer(0.1μg/μL)0.5μL,2.5mM dNTP2.0μL, ddH2O9.4μL.The high/low nitrate content DNA pool was constructed respectively according to the nitrate content. A total of380SRAP markers were screened between two pools. The result indicated that only one difference was found, which was derived from2primers combination.The different fragment was reclaimed from1%electrophoresis gel and cloned through T-easy vector. It showed that the size of inserted bands was same as the target fragment.In this study, the SRAP marker was used to detect marker associated with cucumber nitrate content. Hence, these results of this study provided a theoretical basis for the future quality improvement of cucumber varieties and selecting new varieties.
