Dendrobium huoshanense belongs to the Orchidaceae family, which han beenshown to have immunomodulatory effects. The aim of the study was to investiganteimmunoactivity of different extracts from Dendrobium huoshanense protocorm-likebodies by two extraction methods in order to clarify whether water extract orethanol extract works. The acute toxicity and the mutagenicity of D. huoshanensepolysaccharides have also been evaluated.Water boiled extract, alcohol soluble extract, alcohol precipitated extract andcrude polysaccharide were obtained from D. huoshanense protocorm-like bodies byA extraction method, whose extraction percents were23.0%,10.8%,12.4%and4.3%, respectively. Total alcohol extract, water boiled extract, alcohol solubleextract, alcohol precipitated extract and crude polysaccharide were obtained by Bextraction method, whose extraction percents were23.04%,6.14%,2.74%,3.34%and2.06%, respectively.Different extracts were given to normal mice for30days. After that, the bodyweight of each group of mice increased to different extent and the highest growthrate occurred in the crude polysaccharide group.The experimental results of ConA stimulated splenic lymphocyte of miceshowed that water extract from extraction method A and B showed significantenhancement in ConA-induced spleen lymphocyte proliferation and immunefunction; while ethanol extract had no significant enhancement in immune function.Among all water extracts, the highest enhancement in immune function was thecrude polysaccharide group.The results of acute toxicity test showed that mice administrated with D.huoshanense polysaccharides at the dosage of15.6g/kg·BW were of no death andno significantly toxic symptom during14days. In accordance with the acute toxicstandard, D. huoshanense polysaccharides had no toxicity. The results of Ames testindicated that D. huoshanense polysaccharides had no mutagenicity.In summary, D. huoshanense polysaccharides had immune enhancementfunction, no toxicity and no mutagenicity. The results would provide a theoreticalbase for the development and application of the medicial value of D. huoshanense.
