Using biochemistry, molecular biology, protemics, carry out a series of studiesto heat and salinity stress on the liverã€nierenã€skinã€muscle and blood of turbot.Analyse the changes of antioxidant enzymeã€Isoenzyme pedigree and the express ofimmunological factors, as a best suited to the environments in which the fish are to befarmed.Two-factor-intersecting experiment was designed to evaluate the effects oftemperature(17℃ã€20℃ã€23℃ã€25℃ã€28℃) and salinity(5‰ã€10‰ã€20‰ã€30‰ã€40‰) on the activitiesã€death rate and antioxidant enzymatic activities in the mucusã€bloodã€gill and liver of juvenil Scophthalmus maximus. The results showed that thefish had a good ingestion in the group of17℃ã€30‰and17℃ã€20‰. Bothtemperature and salinity had significant effect (P<0.05)on the activity of SODã€CATã€GP-X. The activity of SODã€CAT and GP-X changed like broken line with theincrease of temperature and relatively higher at20℃and23℃. The activity ofmucusã€bloodã€gill and liver SODã€CATã€GP-X increased as the salinity increased ordecreased from30‰,and had a lower activity at30‰while higher at5‰and40‰.The temperature and salinity interaction had significant effect (P<0.05) on theactivity of SODã€CAT and GP-X.Polyarylamide gel electrophoresis test was used to study the expression of SODã€ESTã€MDHã€GDH isozyme in liverã€skinã€muscle and gill of turbot at13℃ã€20℃ã€23℃ã€25℃ã€28℃and5‰ã€10‰ã€20‰ã€30‰ã€40‰. The results were as follows:the expression of heat and salinity have effects on all isozyme which change thebrightness of isoenzyme pedigree, the activity of SODã€ESTã€MDH isozyme boost upwith the temperature then begin lower at28℃. The temperature has different effect onGDH in different tissue. The activity of SODã€ESTã€MDH isozyme of four tissue andGDH of the gill boost up compared with control group when the salt changed, whiletrail off in liverã€skip and muscle. To use RT-PCR method determined the immune-related factors includingimmunoglobulin M (IgM), hepcidins, lysozyme heat shock protein70, heat shockprotein90,C-t type lectin, Lily-type lectin in liverã€nieren and skin under the conditionof20℃ã€23℃ã€25℃ã€28℃and5‰ã€10‰ã€20‰ã€30‰ã€40‰.
