1.The species-identification of Hypomesus in ChinaHypomesus is a kind of the transplant fishes of the Chiense inland waters,and it’salso a kind of pint-sized commercial fishes.It has already brought favorable societal andeconomical benefits. However, there are different reports on the problem of the mostfishes of Hypomesus we had transplanted is Hypomesus olidus or Hypomesusnipponensis. In this analysis, we adopt the methord of mitochondrion CO1DNAbacoring to identify48samples collected from different places. The bias of nucleotidebase is not obvious,there are18haplotypes in48samples,the number of largest sharedhaplotype is22.The COI sequences of48samples are similarest to the sequences ofHypomesus nipponensis in the Genbank,and the genetic distance is less than0.01,whliethey have larger difference with the sequences of Hypomesus olidus in the Genbank,asthe the genetic distance is more than0.1. According to the results, the most fishes ofHypomesus we had transplanted is Hypomesus nipponensis.2. Genetic structure of Hypomesus based on MtDNA control regionThis study analyzed the genetic structure of the populations of16individuals ofPond smelt based on the sequence-measure of clone. From the result, there are largeinserts and lacking in genes. Heilongjiang jingbo lake groups have the highest A baseand T bases, Jili lake xiaohaizi have the highest C base,the Fuhai reservoir have thehighest G base and the Jili lake xiaohaizi have the highest C+G base.There are144insert and lack of sites,36variation sites; the Wulungu lake dahaizi has the highestconversion: transversion’s value (R)and the Fuhai reservoir has the lowest. The geneticdistance between the groups in Jili lake xiaohaizi and Heilongjiang jingbo lake(so is Xingjiang Buositeng lake) is the largest and between the groups in Fuhai reservoir andXingjiang Buositeng lake is the smallest.Groups in Fuhai reservoir have the highestSingle type of polymorphism and the Xingjiang Bositeng lake(so is the Wulungu lakedahaizi) have the lowest. The groups in Xingjiang Bositeng lake have the highestNucleotide polymorphism and the groups in the Heilongjiang jingbo lake have thelowest.Through the analysis of FST value,between the group in Heilongjiang jingbolake and Erlian Depression/Yunnan Chaihe Reservoir have obvious differences,but theyhave no obvious differences in the other groups.we found that there were no obviousdifferences in the genes of each group.3.Highly polymorphic microsatellite loci from the HypomesusTen polymorphic microsatellite loci from the Hypomesus were identified and theircharacteristics were described. These markers were tested in a wild populations fromHeilongjiang jingbo lake,China, respectively. We have got391transformants,132ofthem has been Inserted by the Hypomesus genome sequences,114of the132havemicrosatellite sequences in them.The number of alleles ranged from5to16with anaverage of8.8per locus. The Mean expected heterozygosity (He) was0.7666in thepopulation from China, Two of the ten locus significant departure fromHardy–Weinberg equilibrium proportions (P<0.05). These markers have beingemployed in population genetic studies on the giant mottled eel in our lab and may bealso useful in studies on other species of Hypomesus...
