Study On Tissue Culture Of Platycodon Grandiforus

Platycodon grandiforus, for families of Platycodon perennial herb, the root for medicine, has the effects of openning the inhibited lung-energy, relieving a cough, eliminating sputum, diminishing inflammation, bringing down a fever, calming, and so on. It is used in the area of medicine, food, view and admire and annually exported to the countries of East Asia,it is a kind of scarce natural plant medicine in today. This paper mainly studied seed budding rate and growing different effects of the potential through the seed of different seed handling way and adding different culture medium.then reseached the multiple shoot clumps of platycodon induction training, a new training and regeneration seedlings transplant.problems. More over the stem tip and different parts of Platycodon grandiforus used to induce stem section of platycodon hairy root were studied and the different results were analyzed and compared.The results showed that used water of40-50℃to soak the seed for14h, then used the combination of silk liquid training (bⅣ) germination rate was best(92%); then after soaking the seed in the water of40-50℃for14h and in the3%KMnO4solution for5h, and used the silk of the training of the liquid combination (dⅣ), germination rate also by more than90%.Meanwhile, in the investigation of the potential seed germination, highest germination potential for58%, the lowest sprout potential was20%, and the average sprout potential was37.94%, there was obvious differences in different treatment combinations, with water of40-50℃for14h after another, with a silk of the training of the liquid seed germination highest potential58%(dⅣ). Tere was obvious difference between different treatments and different cultures, are between. the interaction of treating way and the medium also presented a obvious difference. In this experiment, in the condition of platycodon induction explant of multiple shoot clumps the optimal culture medium for add0.3mg/L of6-BA and add0.05mg/L of NAA MS culture. The most suitable for Platycodon grandiforus seedlings multiple shoot clumps and consider its induction was growing and subculture proliferation of culture medium was adding2.0mg/L6-BA and0.5mg/L NAA MS culture, and the most suitable for root culture medium of Platycodon grandiforus was adding0.2mg/L NAA1/2of MS culture. In the induction of platycodon hairy root, whether it be of platycodon leaf or stem section, with the concentration of NAA increased, induction of hairy root produced rate of different, Low concentration, hairy root in the direct inoculation explant grown on; When the concentration of NAA was high, the dedifferentiation callus were taken shape, and then the white villous root were formed on the surface of explant.
