The Polygonatum odoratum Mill.Druce is a perennial herb,It is delicious and sweet,and has the function of nourishing Yin and producing saliva.At the same time,it can also be used in porridge or cooking.And the jade bamboo has a very high ornamental value,in the garden to do more applications.Successive years of development led to the increasing shortage of jade bamboo resources.In order to solve this problem,in this experiment,the excellent individual plants of ornamental arborvitae cultivated by human before were used as explant materials to carry out tissue culture technology research.In order to optimize the technical system of tissue culture of this strain,and to provide technical support for its industrialized seedling and rapid propagation.In this experiment,the effects of different sterilization time,different parts of materials,different culture media,different combinations of growth regulators and low temperature treatment on the start-up culture of Polygonatum odoratum are studied.Different hormone combination,sucrose concentration,seedling refining time and activated carbon concentration were used for the proliferation and rooting culture of this species.At the same time,the effect of culture substrate on the survival rate of test-tube seedlings is systematically analyzed.The results are as follows:1.Primary cultureAfter 75%alcohol treatment for 10s,0.1%hg continued treatment for 5min is the best sterilization time combination for the primary culture of the rhizome,and the contamination rate was only 14.00%,lower than other treatment and control.The rhizome was the best rhizome and had the highest survival number.The best basic medium is MS medium,and the induction rate was 88.00%.The optimal combination of hormones was 1.0 mg·L-1 6-BA+0.5 mg·L-1 NAA,the differentiation rate is 87.00%,and the plants grow well.After 4?low temperature processing 7 d the jade bamboo rhizome induction rate is significantly higher than that of without low temperature treatment,the induction rate increased by 24.00%.2.Proliferation and cultureMS medium is used as the basic medium for the proliferation of this strain.When 6-BA is combined with NAA,the proliferation medium MS+0.5mg·L-1 6-BA+0.4mg·L-1NAA have the best effect.When 6-BA is combined with IAA,the proliferation medium MS+2.0 mg·L-1 6-BA+1.0 mg·L-1 IAA have the best effect.When sucrose concentration is 30 mg·L-1,the adventitious bud proliferation rate is the highest,reaching 86.00%.Considering the state of proliferation bud,the rate of proliferation and the growth of adventitious bud,the optimum succession time of this strain is 25 d.3.Rooting cultureThe optimum rooting medium is 1/2 MS.When 0.5 mg·L-1 NAA or 0.4 mg·L-1IAA are added,the rooting rate of adventitious shoots is the highest.Rooting can be effectively promoted when the content of activated carbon in rooting medium is 0.5g·L-1.The sucrose content of 30g·L-1 is the most suitable for rooting.4.Seedling refining and transplantingThe highest survival rate is 90.07%after 3 days.The most suitable planting substrate is turf:garden soil:river sand=1:1:1,and the survival rate is 86.00%.
