| Toxoplasmosis is a kinds of epidemical parasitic disease that is caused by Toxoplasma gondii in worldwide. Toxoplasma gondii, as an obligate intracellular parasite, it can infect humans and all other warm-blooded animals and bring diseases and deaths. Toxoplasma gondii infection not only threatens humans’health, but also causes great economic losses for animal husbandry. At present, the population of keeping the dogs in increasing number, chicken and its products, which all can be the source of infection for human, so it has a great significance on establishing a rapid, reliable diagnosis of Toxoplasmosis and making the epidemiological investigation in time. Serologic test is one of the methods commonly used in diagnosis of Toxoplasma gondii. Modified agglutinate text (MAT), one of serologic test, is widely used in the large-scale Toxoplasma gondii infection epidemiology investigation and research in European and American countries because of its simple, quick, and the characteristics of high sensitivity and specificity, however, related reports using this method are lack in our country.Modified agglutination test was developed to detect Toxoplasma gondii antibodies, which requires large amounts of antigen of Toxoplasma gondii antibodies. The antigen is prepared using the RH strain of toxoplasma cultivated along with mouse S180sarcoma cells in the peritoneal cavities of mice. Toxoplasma gondii tachyzoites were purified and suspended in6%formaldehyde solution, The parasites are then suspended in an alkaline buffer pH8.9. Sera was diluted by0.2M β-ME PBS, so as to establish the MAT and improved the specificity and sensitivity of MAT.20positive serum of rabbits and10negative serum of mouse were detected by MAT, the results show that coincidence rate was100%. Seroepidemiological survey of Toxoplasma gondii infection in both925dogs that from Shandong, Jiangsu, Hunan and Hubei Provinces, and610chicken from Jinzhou were detected by MAT, and indirect hemagglutination test(IHAT) as a comparison. The results showed that on the MAT, IHAT dog Toxoplasma gondii disease detection rate were8.22%(76/925),2.05%(19/925), both very significant difference (P<0.01); MAT, IHAT to the chicken Toxoplasma gondii disease detection rate were9.02%(55/610),1.64%(10/610), both very significant difference (P<0.01).Seroepidemiological survey of Toxoplasma gondii infection in dogs and chicken were detected by MAT. The result show, positive rate in Shandong, Jiangsu, Hunan and Hubei were13.5%(27/200),2%(4/200),1.54%(5/325),20%(40/200); positive rate in male and female were7.98%(36/451)and8.44%(40/474), it was no significant deviation in different gender (P>0.05). The infection rate of Toxoplasma gaondii in the chicken from Jinzhou,55out of the610chicken contained specific antibodies to Toxoplasma gondii(9.02%). The positive rate in chickens free range and fed in cages were18.75%(30/160) and5.56%(25/450), there was a significant difference in two(P<0.01). The commercial layer’s infection rate was7.89%(15/190), the infection rates in commercial broiler and breeder were1.25%(2/160) and8%(8/100). The positive rates of serum antibody among3kinds of chicken were significant difference(P<0.05).In conclusion, this was the first study on epidemiological survey of Toxoplasma gondii infection by modified agglutination test in dogs and chicken in parts of China, it provide a method for epidemiology investigation of Toxoplasma gondii infection. It was hoped that the result can be a reference in prevention of Toxoplasmosis. |
