This experiment was conducted to study the effects of different functional oligosaccharides combination on rumen micro flora of Jinjiang cattle. Three healthy and similar body weight Jinjiang cattle with permanent rumen cannula were selected, the test is divided into three periods, the first period:the control period, fed the basal diet without any oligosaccharides, the second period,GTl period,fed the basal diet+0.8%MOS+1%FOS+0.8%SBOS,the third period:GT2period, fed the basal diet+0.8%MOS+1.2%FOS+1%SBOS,experiment lasting14days of each period. This study was comprised of3sections:(1) Using the nylon bag method, study the effects of functional oligosaccharides combination on rumen degradability of rice straw of Jinjiang cattle. The rice straw degradability of DM, OM, CP, NDF, ADF after72h on rumen of Jinjiang cattle was determined, and then analysised the correlation. The results showed that:compared with the control period, GT1period and GT2period were very significantly improve the degradabi-lity of DM,OM,NDF,ADF(P<0.01), and difference between the GT1and GT2period was not significant (P>0.05). The degradability of CP was not significant (P>0.05) between each period. The correlation of rumen degradability after72h between DM、NDF、ADF to OM of Jinjiang cattle is strong, the rumen degradability of CP was not closely related to OM.(2)Study the effects of functional oligosaccharides combination on rumen solid bacteria diversity of Jinjiang cattle by PCR-DGGE technique. The results showed that:the effects of adding functional oligosaccharide combination on rumen solid bacteria of PCR-DGGE fingerprints bands of Jinjiang cattle were not obvious, Similarity degree of three trial cattle was high in three perods.(3) Study the effects of functional oligosaccharides combination on rumen liquid bacteria diversity of Jinjiang cattle by PCR-DGGE technique, and then specific bands were cloned and sequenced. The results showed that:after adding functional oligosaccharides combination, the bands which analyzed by PCR-DGGE increased significantly on rumen liquid bacteria of Jinjiang cattle, and increases the diversity of rumen microflora, GT2period is better than GT1period. Adding functional oligosaccharides combination produced12specific bands. By sequencing,2strains belongs to Fibrobacter succinogenes, the other10strains in gene bank belongs to uncultured bacteria. Seen from the phylogenetic tree, these12strains belongs to Eubacterium, Fibrobacter succinogenes, Clostridium, Succiniclasticum, Prevotella and uncultured rumen bacteria. Among them, GT2period produced Prevotella and Fibrobacter succinogenes coexistence can exponentially improve the digestibility of hemicellulose and pectin, which better than GT1period. The results indicated that adding functional oligosaccharide combination can beneficial to fiber degradation and utilization of Jinjiang cattle, improve rumen microflora. Comprehensive analysis, GT2period is better than GT1period, recommended to supple with0.8%MOS+1.2%FOS+1%SBOS functional oligosaccharide combination in the diets of Jinjiang cattle.
