The study was conducted to investigate the dynamic changes of soluble proteinand free amino acids in fluid of chicken embryo. A total of160White Featherbroilers (Exp.1) or Three Yellow chicken (Exp.2) eggs were divided into8treatments (sampling points). Twenty eggs were sampled at day6,7,8,11,13,14,16and18for analysis of soluble protein content in allantoic and amniotic fluids, and freeamino acids concentration in allantoic, amniotic and yolk fluids.The results in White Feather broilers showed that:(1) The soluble proteincontent in allantoic fluid was always increased. The insoluble protein content inamniotic fluid increased significantly before16d, but decreased significantly soonafterwards (P<0.05).(2) All amino acid concentrations in allantoic fluid appeared apeak in7d, and always increased in13-16d, but Asp, Ala, Met, Ile, Leuconcentration decreaseds significantly during the late stage of incubation (P<0.05).All amino acid (especially Arg) concentrations in amniotic fluid were alwaysincreased. All amino acids concentration were increased at first and then decreased,and the peak appeared in11d. The results in Three Yellow broilers showed that:(1)The soluble protein content in allantoic fluid was always increased, which is similar tothat in White Feather broilers. The soluble protein content in amniotic fluidsignificantly increased before16d, but decreased significantly (P<0.05) soonafterwards, which is also similar to that in White Feather Broilers.(2) All amino acidconcentrations in amniotic fluid began to increased in7d. All amino acids (exceptAsn) increased significantly in14d (P<0.05), but Asp, Gln, Arg and Pheconcentrations were decreased significantly during late stage of incubation (P<0.05).;All amino acid concentrations in allantoic fluid appeared a peak at7d, and wereincreased during late stage of incubation. After14d, most amino acid concentrationswere decreased significantly in yolk fluid (P<0.05).These results suggest that solubleprotein content the amniotic fluid is negatively correlated with the chickenembryo growth. The Gln and Arg had higher concentrations in amniotic fluid.Allantoic sack was the store for Arg.
