Salvia miltiorrhiza with important value are Chinese traditional medicine materials for exportation,80%of which are wild resources that are overly excavated and reduced day and day. And there are a lot of problems for manpower planting, such as no good seeds, quality degeneration, long production period, seed unhealthiness, excessive rudimental pesticide, high difference for the composition and content of effective component, and the unstability for clinical treatment.However, the technology of tetraploid induction and hairy root culture can produce a large number of salvia miltiorrhiza materials, improve its quality, and greatly enhance its medicinal composition quantity, quality and stability. So according to the problems for salvia miltiorrhiza planting, clinical treatment and its continuable development,the study employed tetraploid induction and hairy root culture technology to study salvia miltiorrhiza,which was favorable for well-bred seeds, advancing medicinal composition, industrialization of salvia miltiorrhiza resource on a large scale, exploring a new path for salvia miltiorrhiza resource exploitation, and the same time supplying theory foundation and technology support for protecting salvia miltiorrhiza resource and biology diversity. The main results were as follows:1、The aim was to explore the feasibility for colchicine to induct Shandong Salvia miltiorrhiza shoots. Result was that the highest induction rate of tetraploid,36.6±0.49%was obtained from the solid medium supplemented with12mg·L-1colchicine. But its survival rate is only16.7±0.670%. Adding8mg·L-1colchicine, the polyploid induction rate was15.9±0.897%, the survival rate of up to40±0.84%. Shandong salvia miltiorrhiza polyploid plants has thick, large and dark leaves. Leaf stomatal length on the lower epidermis of leaves was long, but its density was small. colchicine can be effective in inducing multiple shoots. And Salvia miltiorrhiza polyploid breeding material was obtained, laying a basis for shortening breeding process.2、The expriment established Shandong Salvia miltiorrhiza hairy root culture system. And cultured hairy roots comed from the same hairy root. So they had inheritance stability, which supplied unified materials for the next test.3、The expriment studied effects of different exogenous hormone on the hairy root growth and biosynthesis of tanshinone ⅡA in Shandong Salvia miltiorrhiza. Results realized that the combination of GA and6-B A at different concentrations adding to1/2MS medium caused an obviously inhibitory effect on the hairy root growth of Shandong Salvia miltiorrhiza. And the highest inhibition rate was up to100%(hairy roots were death). However, the growth of hairy roots could be obviously activated by the combination of KT (1.0mg·L-1) and IBA (1.0mg·L-1), increasing by3.251folds compared with the control. Total tanshinone ⅡA content in hairy roots bodies is the highest under the combination of3.0mg·L-16-B A and0.2mg·L-1NAA.6-B A of2.0mg·L-1played a significant role in promoting tanshinone ⅡA biosynthesis, and the tanshinone ⅡA concentration was3.012times higher than the control. Different exogenous hormones and their combination adding to1/2MS medium had significant influence on the hairy root growth and the content of tanshinone ⅡA in Shandong Salvia miltiorrhiza.4、The expriment studied effects of different pH value on the hairy root growth and biosynthesis of tanshinone ⅡA in Shandong Salvia miltiorrhiza. Results realized that the pH value of9could make the hairy root growth and the content of tanshinone ⅡA get up to the highest quantities in Shandong Salvia miltiorrhiza.
