Salvia miltiorrhiza is an important medicinal plant that has many effects such as blood circulation disturbance, diminishing inflammation and analgesia, blood pressure-lowering, etc. It is a safe and reliable natural Chinese medicine for treatment of cardiovascular disease. In the increasingly clarity of wild Salvia miltiorrhiza’s medicinal and healthcare function, its demand is increasing year by year. However, excessive harvesting of Salvia miltiorrhiza has changed its demand from bad to worse. Although, the steady increase on area and yield of cultivated Salvia miltiorrhiza has alleviated these problems to a certain extent, which exists some serious problems such as low active ingredient, long growth cycle, degradation of quality. Above all, the basic and applied work in scale industrial production of Salvia miltiorrhiza effective secondary metabolites by hairy root gains more and more attention.Hairy root technology culture is a plant transformation techniques which developed on the basis of plant cell culture technology. It inserts Agrobacterium’s Ri plasmid gene fragment into plant genome, so plants produce large amounts of hairy root. Hairy roots hold the advantages of genetically stabilization, high ingredient’s content and cultivation process without hormones and so on. Technology of Salvia miltiorrhiza’s hairy root still exists some problems, such as the uncontrollable quality and the low content of secondary metabolites.In order to solve these problems, this paper carried on the material of hairy root, the time of fermentation and the effect of elicitor. The aim of research is to screen out fine strains of Salvia miltiorrhiza, make sure the optimum fermentation period and enhance the Salvia miltiorrhiza’s hairy root capacity of effective secondary metabolites by elicitor. The main contents and results are as follows:1. In view of 7 fine strains of Salvia miltiorrhiza D5, D7, D20, D21, T230, T310, T810, an effective system of hairy root induction and fermentation has been established. We compare induction efficiency, growth rate, content of active component from different genetic background Salvia miltiorrhiza’s hairy root, we screen out D5, D20, and D21 as the superior Salvia miltiorrhiza strains.2. Dynamic of secondary metabolites has been determined in different period of Salvia miltiorrhiza’s hairy root fermentation system. The law of accumulation of Salvia miltiorrhiza’s hairy root secondary metabolites has been explored. We compare the content of four secondary metabolites of Salvia miltiorrhiza’s hairy root, which ferments 20 d, 40 d, 60 d, 80 d, 100 d respectively. We determine 80 d as the optimum fermentation period of phenolic acid and 100 d as the optimum fermentation period of diterpenoid compound.3. We test whether the elicitor of methyl jasmonate has an effect on the content of secondary metabolites of Salvia miltiorrhiza’s hairy root. The aim is to make sure the effect of methyl jasmonate. The results show that Rosmarinic acid and salvianolic acid B are increased by 1.34 times and 4.34 times respectively, A diterpenoid tanshinoneⅠand tanshinoneⅡA are increased by 1.49 times and 1.13 times respectively.
