Astraglus and Acanthopanax as herb were used in traditional Chinese medicinefor thousand of years in modulating immune status of human and relievingimmunosupression on animal. In this study, a formula has designed to test themodulating safety and efficacy on enhancing immune response on7d old chicks,which consists of Astraglus and Acanthopanax, named as Qiwujia Soluble Powder(QSP). The results will provide evidence for its further clinical application.1. Clinical Safety Trial: Chicken were treated with QSP for continuous7days at the dose of,1.2,2.0and4.0g/kg body weight after melting by warm water in the morning and afternoon. Nomorbidity, mortality and other toxic reaction were observed. All above resultssuggested that QSP has higher safety in chicken.2. Clinical Preliminary Trial: To determine the effective dose and effective doserange of QSP for clinical trial, healthy Hy-line brown chicken as target animal wereattenuated and inactivated oil seepage vaccine immunity by Newcastle disease and treated withQSP(0.05,0.10.0.20,0.40,0.80,1.60g/kg). The results showed that QSP own a betterimmune enhancing effects, which could increase the serum antibody titer ofNewcastle disease virus, promote peripheral blood lymphocyte proliferation,improve the rate of erythrocyte C3bR yeast rosette, increase the indexes of thymus,spleen and bursa, and improve weight gain and reduce the feed-meat ratio. Wefound that0.4g/kg was the optimal effective dose, and0.2-0.8g/kg was the effectivedose range.3. Clinical Trial: The1350healthy male Hy-line brown7-day-old were treatedwith two methods. The results showed that QSP could significantly increase theimmune function in chicken, and own the function of improving weight gain andreducing the feed-meat ratio. QSP could significantly increase the serum antibodytiter of Newcastle disease virus, promote proliferation of peripheral bloodlymphocyte, improve the rate of erythrocyte C3bR yeast rosette and the phagocytic index of mononuclear-macrophages, increase the content of IFN-γ, decrease thecontent of IL-4, increase the indexes of thymus, spleen and bursa. In addition, QSPcould against the immunosuppression induced by hexadecadrol. The dose of0.4g/kgbody weight is recommended for clinical medication with3days before vaccinationfor continuous5days.4. Enlarge Clinical Trial: Fifteen thousands six hundred and seventy healthyHy-line brown7-day-old were divided into three groups in the chicken farm.Theseresults indicated that QSP could significantly increase immune function in chicken.The dose of0.4g/kg body weight is recommended for clinical medication. Theresults showed that QSP could significantly improve the antibody titer level ofNewcastle disease virus, promote proliferation of peripheral blood lymphocyte,increase the rate of erythrocyte C3bR yeast rosette, and reduce the morbidity andmortality to some extent.In summary, QSP has higher safety in chicken and can enhance the immunefunction of chicken.0.4g/kg body weight is recommended for clinical medicationwith3days before vaccination for continuous5days. These results provide evidencefor clinical application of QSP.
