Phylogenetic Relationship Analysis And Fingerprinting Of Pigmented Potato With SSR Markers

Potato, as very important food and vegetable crop in the world, is more prominent increasingly. Pigmented potato which are rich in Anthcyanins have more nutritional value. For the purpose of germplasm identification at the molecular level, SSR markers were employed to analysis on genetic diversity and fingerprinting of86pigmented potato genetypes. The main results are as follows:1. Fifty-six pairs of SSR primers showing polymorphism were selected by screening over200SSR markers for analysis on phylogenetic relationship analysis and fingerprinting.2. Fifty-six primer pairs amplified a total of238alleles(including232polymorphic alleles)among the86genetypes, and the ratio of polymorphism was as high as97.48%. Alleles amplified by each pair of primer ranged from1to11, with a mean of4.25. The polymorphic information content values(PIC) ranged from0.3253to0.9394, with a mean of0.7985.The fragment sizes varied from79bp to318bp.3. UPGMA cluster analysis showed that all the cultivars could be assigned into three proups at genetic similarity of0.63. The genetic similarity between all the materials except JK2are higher than0.6, and some materials share the same bands in every alleles. It’s indicated that pigmented potato cultivars have a narrow genetic basis.4. Five pairs of primer which were S038、S072、S102、StI005and StI007could distinct all the86materials, and were used to establish DNA fingerprints for the86pigmented materials which were clarified as80genotypes.5. Content of carotenoid、vitamin C、chlorogenic acid、total phenols and solanine in tubers of some pigmented potato genotypes were measured in this study.
