Photosynthesis and some other physiological and biochemical characteristics are directly bound up with growth, development and output of plants. Investigation of the photosynthesis characterization of transgenic plants is benefit to explain the change of genetic modification plants nonscheduled photosynthesis. In this study, the pot experiments of barley were carried out. The experiment result of the transgenic barley and original barley varieties were analyzed. The scale characteristics of photosynthesis as well as the characterizations of reflectance spectral and photosynthesis-related protein atlas of the T1and T2generation transgenic barley photosynthesis were studied via a portable photosynthesis system LI-6400XT, a field portable spectrometer Unispec-Sc and SDS-PAGE electrophoresis.The results showed that for the scale characteristics of photosynthesis, the parameters, such as net photosynthetic rate, transpiration rate, stomatal conductance, energy efficiency and water use efficiency of the T1generation transgenic barley during tillering and heading stage have no significant difference between their respective control. The differences of the photosynthetic parameters mentioned above between different genotypes are also not notable. In addition, stomatal conductance and net photosynthetic rate, the transpiration rate and stomatal conductance, net photosynthetic rate and transpiration rate showed a good linear relationship. For the aspect of reflectance spectral characteristics, the difference between spectral reflectance values of GM and Non-GM barley mainly concentrated on the Red Valley zone with a wave length range from650to700nm and the Green Valley zone with a wave length range from400to500. The difference in change between reflectance spectra of the GM and Non-GM barley, such as NDVI, Chl NDI, ARE, PRI, RRED/RGREEN and NIRR800during tillering and heading stage showed different characteristic which has some relationship with the barley lines. For the aspect of photosynthesis-related protein atlas, the content of soluble protein and thylakoid membrane proteins of different strains of transgenic barley of T1generation during tillering and heading stage showed no significant difference between the respective non-transgenic controls. The significant change of the content of soluble protein and thylakoid membrane proteins of different strains of transgenic barley mainly depended on the development stage and barley genotypes. During the tillering stage the soluble protein and thylakoid membrane proteins profiling of different barley exhibited no significant change. However, during the heading stage the soluble protein and thylakoid membrane proteins profiling of transgenic barley lines showed a significant change. The subunit types of changes are relevant to the barley lines.
