Duing the plant process of ginseng, ginseng was easily infected by ginseng pathogenic bacteria, so that the ginseng would be sick, the quality and yield of ginseng would be influent. For this reason every year economic loss was enormous, so that how to ensure to decrease the infestation of plant disease during the normal growth of ginseng has been the research focal point of ginseng plant personnel and scientific research personnel for the past many years. Although the application of traditional chemistry pesticide could have precautionary and inhibitory action, along with the deep realization of chemistry pesticide and its perniciousness during its long-term use, chemistry pesticide was necessarily substituted with biological pesticide, which had little impact on health and environment.This text aimed at current situation, to begin with plants and Microorganisms bacteriostatic two respects, we screened the traditongal Chinese medicine and fungi which had inhibitory action to ginseng pathogenic bacteria, we carried on initial study.1、We got113strains germ, which was separated from the soil samples in gingeng plant south district of JiLin, fungi23strains, bacteria66strains, actinomycetes24strains, bacteria PFD2had good inhibition action to ginseng dark spot and gray mold pathogenic bacteria, so we took this bacteria as important research object and carried out deep study. We could identify that bacteria PFD2was bacillus from morphology, we couldnot exactly decide its classification status. We adopted16SrDNA sequence analysis to assay, which showed it had high similarity with bacillus polymyxa, and we finally determined this strain was bacillus polymyxa, named JL2011.We optimized and choosed fermentation condition and fermention medium of the strain PFD2. Firstly, we adopted single factor test to dicide the best carbon source was glucose, the best nitrogen source was peptone. We carried on contrast test of nutritive material addition by orthogonal test, and decided the best fermentation condition and culture medium:glucose2%, peptone0.3%,0.5%NaCl, the best temperature was28℃, cultivation days was2days, pH was6, the volume of liquid was100mL/250mL, the inoculum size was4mL, the rotation speed was140r/min.2、We choosed the ethanol and water extracting solution of Radix Glycyrrhizae, Fructus Cnidii, Semen Plantaginis, Herba Agastaches Rugosae, Radix Puerariae Lobatae, Cortex Cinnamomi Cassiae six traditional Chinese medicine, and adopted mycelial growth method to choose bacteriostatic activity to ginseng gray mold pathogenic bacteria, ginseng rust rot pathogenic bacteria, ginseng black mold pathogenic bacteria and ginseng stem rot pathogenic bacteria, the result was the bacteriostatic activity80%ethanol extracting solution of Radix Glycyrrhizae, Cortex Cinnamomi Cassiae was obvious, the extracting solution of Cortex Cinnamomi Cassiae had the highest bacteriostatic activity, the effect was up to100%to ginseng rust rot pathogenic bacteria and ginseng dark spot pathogenic bacteria, the effect was inferior to gray mold pathogenic bacteria and stem rot pathogenic bacteria.3、The strain PFD2fermentation liquid and extracting solution were both used for ginseng, we measured the inhibition. The different Radix Glycyrrhizae and Cortex Cinnamomi Cassiae addition and the strain PFD2cofremented, we measure bacteriostatic activity effect of fermentation of supernatant, by study, it showed that after cofermentation,, the growth of strain PFD2was influent, and the bacteriostatic activity fermentation liquid was not well. The traditional Chinese medicine ethanol extracting solution and the strain PFD2fermentation supernatant mixed and were used to measure bacteriostatic activity effect, the result showed V fermentation:V Cortex Cinnamomi Cassiae=5:3had best bacteriostatic activity effect.
