Helicoverpa armigera (Hübner) and Helicoverpa assulta (Guenée) are bothbelong to Lepidoptera, and important pests on peppers. However, hosts of H. assulta areless than H. armigera, H. assulta just feeds on a few kinds of plants, like tobacco, pepperand so on. In addition, many studies show that H. assulta are more likely to feed pepperthan H. armigera. After eatting artificial diet added different kinds of pepper, theexpression of β-fructofuranosidase, Heat shock protein Hsp70and eukaryotic initiationfactor5A in labial glands of H.armigera showed significantly different.After eatting artificial diet added different varieties of pepper, to prove the differencesof the three proteins on the level of gene transcription in the two noctuids, reveal themolecular mechanisms about degrees for suitable host of the two noctuids on differentpepper varieties, open new avenues for biological control of the two noctuids on pepper. Inthis study, contrast with normal artificial diet, adding No.2Yuyouxianla, Yuyilüguan,Yuyimoxiudajiao and No.12Yutianjiao pepper varieties in which the content of capsaicinreduces in turn, to feed the end of4th instar larvae, the total RNA of the labial glands ofthe two noctuids was extracted after0h,24h,48h,72h and96h, then RNA was reversetranscripted to cDNA. Real-Time PCR was used for analyzing the difference ofβ-fructofuranosidase, hsp70and eIF5Aon the relative expression level. The main results asfollows:1. The treatment of higher capsaicin could promote the β-fructofuranosidaseexpression of H. armigera but inhibit the gene expression of H. assulta; the treatment oflower capsaicin could promote the β-fructofuranosidase expression of H. assulta but inhibitthe gene expression of H. armigera. The high level expression of β-fructofuranosidasegene with a short time after the treatment of No.2Yuyouxianla containing the highestcapsaicin in H. armigera, but which after the treatment of Yuyimoxiudajiao and No.12Yutianjiao containing the lower capsaicin. The expression rule of β-fructofuranosidase wasconsistent with the fitness difference of H. armigera and H. assulta on different peppervarieties, presumably the gene expression of the two moths was in connection with thefeeding adaptation mechanisms on different pepper varieties.2. Continuous exposure to four pepper treatments could inhibit hsp70expression in H. armigera, but in H. assulta put up promotion to varying degrees. Whether the same timeon different treatment, or the same treatment at different time, both present the expressiontrends of hsp70in H. armigera was complex, should ever start repeating; but in H. assultaoperated steady changes on four treatments, along with the extension of treatment time, therelative expression level gradually raised, reached the highest at96h. The protein Hsp70isclosely related to the tolerance ability of biological resistance pressure on theenvironmental stress, the analysis of differential expression of hsp70in the two moths ondifferent pepper treatments laid a foundation for the function research of capsaicin stressaspects.3. The expression of eIF5A on four treatments were subdued in H. armigera, but werepromoted in H. assulta to a varying extent, and with the decrease of capsaicin, the genepromote time gradually put off. Whether the same time on different treatment, or the sametreatment at different time, the expression trend of eIF5A was in keeping with hsp70in H.armigera and H. assulta. The expression change trend of the gene was complex in H.armigera, while present fewer changes on four treatments in H. assulta, with theprocessing extension of time, the expression quantity has been on the rise, and prepared thehigh expression level at96h on the treatment of Yuyilüguan and No.12Yutianjiao. Thestabilization trend of the expression of eIF5A, implys that H. assulta is more tolerance tocapsaicin stimulation than H. armigera, suggest the gene transcription expression wasneeded for some special mRNA translation, which played an important role in resistance tocapsaicinoids stress.
