The experiment was conducted to a new epidemic attacked on the ducks and which can cause the ducks to get a sharp decline-named as duck hemorrhagic ovaritis, DHO. The main purpose of this experiment is to conduct the pathological analysis of the pathological changes of duck hemorrhagic ovarian minflammation incidence duck antigens in the body distribution, and to create artificial infection model duck hemorrhagic ovarian inflammation on different days of age ducks, and also provide a reliable reference for advice on disease prevention and control for future.Test methods:In this experiment, we used the normal negative production ducks as the object of study, and used the DHO-F4generation of virus solution which was breeded by the Institute of Animal, belongs to Beijing Academy of Agiculture and Forestry Sciences, to attack ducks. After that, we should observe and record the incidence of the ducks. After each batch of ducks were inoculated in the first4,5and6days, one or two duck in each group would be randomly seleted for hunting, blood collection and separating the serum. At the same time, we should take throat swabs and rectal swabs from the attacked ducks to inoculate chicken embryos, and observe the chick embryo deaths.And we should autopsied corpse, derived the tissues and organs Systematically and fixed them with formalin. After that we could observed the HE slices from the Pathological point of view. At the same time, we needed to do immunohistochemistry for antigen localization and detection.The results of the experiment:the artificial infection moded about350and500days of age ducks were successfully created with DHOV. The attacked ducks manifested in the follicle degeneration, deformation, bleeding, enlarged spleen, mild brain edema and other characteristic lesions, those were all fit the natural case characteristics. The immunohistochemical results showed that the positive reaction appeared in the microglia of the brain, spleen lymphocytes, the liver cells from the liver, kidney tubular epithelial cells and theca cells. And the amount of the virus attacked the ducks was greater, the pathological phenomenon was more obvious, also the positive immunohistochemical reaction was stronger.The conclusion of the experiment:duck hemorrhagic ovarian could be successfully infected ducks by artificial means. And the virus could cause the ducks appeared not only hemorrhagic ovarian inflammation but also the organizations and organs characteristic lesions such as intestitial hyperplasia after inoculation. The immunohistochemical results showed that the antigen was found primarily in brain, liver, kidney, spleen, ovaries and other parts.
