Choose Trichoderma viride as material,study intensively on its metabolites.Include the research on its antifungal activity, medium of optimization,metabolitessstability,isolation and purification of metabolitess,antifungal activities ofcompounds.We need to develop a kind of steady efficient and safe biologicals tofright postharvest diseases of fruits,in stead of chemistry germicides.The study on the utilization ratio of carbon source and nrogen source,showedthat about carbon source,Trichoderma viride had highest utilization ratio ofgalactose,lowest utilization ratio of sucrose and moderate utilization ratio of lactose,trehalose,xylose,fructose,mannose,glucose;about nitrogen source,Trichoderma viridehad higher utilization ratio of organic nrogen source and amino acid than inorganicnitrogen source.To optimize the cultural conditions of metabolites of Trichoderma viride,response surface method (RSM) were used.On the basis of the previous results andaccording to the central composite experimental design principles,the responsesurface method with3factors and3levels was adopted.The results showed that theoptimum conditions were that the potatoes content of20%,fermentation days of4.5dand the volume of liquid of67mL/mL.Under these conditions,the predicted value ofthe antifungal activity could be up to81.89%,in basic agreement with the actualvalue of83.03%.The absolute error is1.13%.On the basis of optimized cultural conditions,study on the metabolites ofantifungal activity of Trichoderma viride to Rhizopus Aspergillus nigerColletotrichum gloeosporioides Colletotrichum gloeos Fusarium oxysporumPenicillium griseofulvum,and its stability.Results showed that the metabolite hashighest antifungal activitiy to Colletotrichum gloeosporioides,up to84.32%;it wasstable to light heat and microwave;be stable when pH6-7; Cu2+Fe3+have large effeton its stability Preservative oxidizer and reducing agent have unsignificant effectwithin the scope of the concentration.The metabolite of Trichoderma viride were extract by different solvent,and theresult showed the antifungal activities of the solvent extracts were different.petroleum ether extract had strongest antifungal activities,then ethyl acetateextract,and n-butyl alcohol was the weakest.Twenty-nine compounds were isolated from the petroleum ether extract of themetabolites through GC-MS,and five of them were determined,there were2,3-dimethylpentan-2-ol Eucalyptol Phenylethyl alcohol Diisobutyl phthalateSohirnone B.Three compounds were isolated from the ethyl acetate extract of themetabolites of Trichoderma viride through repeating column chromatography onsilica gel,sephadexLH-20and determined by chemical and modern spectroscopicanalysis,which include Ethyl sorbate Bisvertinolone Viridenepoxydiol.The last the antifungal activity of the pure compounds were studied on.The resultshowed that Ethyl sorbate and Bisvertinolone had antifungal activity toColletotrichum gloeos,but Viridenepoxydiol hadn’t;and Bisvertinolone MIC50was5.95mg/mL.
